摘 要:为优化穿山甲的分子鉴定方法,采用9种穿山甲54条Cyt b基因完整序列,评估了MEGA11软件中的7种模型和23种碱基替换类型所估算的遗传距离的物种分辨率、Cyt b基因全序列及各区域鉴别力。结果表明:K2P模型和p-distance模型基于颠换(T_(V))计算的遗传距离具有最优的物种分辨率;Cyt b基因全序列有较好的物种鉴别能力,其中,第301~400号和第851~950号的2个碱基序位片段组合时具有最高的物种分辨率,是鉴别9种穿山甲的最佳理论区段。现有的方法中推荐使用Kocher等的引物。今后在研发中采用多重PCR结合高通量测序对多片段组合进行分析,将会提高穿山甲的鉴别力。In order to optimize the molecular identification of pangolins,we evaluated species resolution of genetic distance estimated using seven models and 23 nucleotide substitution modes in MEGA 11 and the power of species identification of the whole and segments of Cyt b gene using 54 complete Cyt b sequences of nine pangolin species.The results showed that the intra-and inter-specific distance computed using K2P and p-distance model with transversion(TV)as substitution mode posed the greatest species resolution.The whole sequence of Cyt b gene had acceptable species identification power,but segments ranging combination between the 301^(st)~400^(th) and 851^(st)~950^(th )nucleotide showed optimal species identification power,is the best section for identifying nine species of pangolins.Among the existing methods,it is recommended to use the primers of Kocher et al.The above results suggest that use of multiple optimal segments would substantially improve the identification accuracy of pangolins given multiplex PCR and massive parallel sequencing approach are used.
