摘  要:以彩色冬小麦TW10-1154、太黑3号、太蓝1号、张紫麦1号、穗选5号、冬黑10号、农大3153和西农黑等8个品种为试验材料,采用荧光原位杂交(FISH)技术以pSc119.2(绿色)和pTa535(红色)为探针,明确8个彩色冬小麦均属于六倍体材料(AABBDD,2 n=42)。同时,对这8个彩色冬小麦进行常温处理(25℃)和冷胁迫(4℃)处理,并结合NBT组织化学染色法进行ROS定位。结果表明,冷胁迫处理下,张紫麦1号、冬黑10号和西农黑的长势最好,在根尖顶端组织区域ROS信号富集最多,而在伸长组织区域ROS信号随着胚根的生长而逐渐减少,因此,推测ROS信号可能是一种动态传递的信号分子。对冷胁迫下成熟期8个供试材料的根系进行拍摄,结果发现,太蓝1号、张紫麦1号、穗选5号、冬黑10号根须健壮发达,表明这4个品种的小麦生长能力和抗冻性强,适宜陇东地区种植。Using colored winter wheat(Triticum aestivum L.)TW10-1154,Taihei No.3,Tailan No.1,Zhangzimai No.1,Suixuan No.5,Donghei No.10,Nongda 3153 and Xinonghei were selected as experimental materials,fluorescence in situ hybridization(FISH)technique was conducted,using probe pSc119.2(green)and pTa535(red)as probes to confirm all the 8 colored winter wheat as hexaploid materials(AABBDD,2 n=42).At the same time,the 8 samples of colored winter wheat were treated with normal treatment(25℃)and cold stress(4℃),and the ROS localization was performed by NBT histochemical staining.The results showed that under cold stress(4℃),the growth of Zhangzimai No.1,Donghei No.10 and Xinonghei was the best,and the ROS signal was most enriched in the apical tissue region of root tip,while the ROS signal in the extended tissue region was gradually reduced with the growth of radicle.Therefore,it was suggested that ROS signal might be a dynamic signaling molecule.The root system of 8 tested materials at maturity stage under cold stress was photographed.The results showed that the roots of Tailan No.1,Zhangzimai No.1,Suixuan No.5 and Donghei No.10 were robust and developed,indicating that these 4 varieties had strong growth ability and frost resistance,and were suitable for planting in Longdong region.
