摘  要:以大桥虎耳草组培苗的叶片为外植体,研究植物生长调节剂对叶片愈伤组织和不定芽诱导、不定芽增殖及丛芽生根培养的影响,进一步探讨两种生根方法对幼苗生长的影响,建立了完善的离体培养体系和植株再生体系。结果表明,大桥虎耳草组培苗叶片愈伤组织和不定芽诱导的培养基为1/2 MS+6-BA 2.0 mg/L+NAA 0.2 mg/L,1/2 MS+KT 1.0 mg/L+NAA 0.2 mg/L和1/2 MS+TDZ 0.4 mg/L+NAA 0.2 mg/L,愈伤组织诱导率达97.78%以上,芽诱导率达96.67%以上,丛芽诱导率达87.04%以上;不定芽增殖培养基为1/2 MS+6-BA 1 mg/L+NAA 0.2 mg/L,芽增殖系数为4.03;丛芽生根培养基为1/2 MS+IBA 0.5 mg/L,生根率为100%,生根条数为10.81(组培苗可瓶外生根)。组培苗移栽后生长良好,移栽成活率达95.56%以上。Using the leaves of the tissue-cultured seedlings of Saxifraga daqiaoensis were taken as explants,the effects of plant growth regulators on callus and adventitious bud induction,adventitious bud proliferation and root culture of the leaves were studied.The effects of two rooting methods on seedling growth were further discussed,and a complete in vitro culture system and plant regeneration system were established.The results showed that the culture-medium for callus and adventitious bud induction were 1/2 MS+6-BA 2.0 mg/L+NAA 0.2 mg/L,1/2 MS+KT 1.0 mg/L+NAA 0.2 mg/L and 1/2 MS+TDZ 0.4 mg/L+NAA 0.2 mg/L,Callus induction rate was over 97.78%,bud induction rate was over 96.67%,and bundle bud induction rate was over 87.04%.The culture medium of adventitious bud proliferation was 1/2 MS+6-BA 1.0 mg/L+NAA 0.2 mg/L,and the bud proliferation coefficient was 4.03.The rooting medium was 1/2 MS+IBA 0.5 mg/L,the rooting rate was 100%,and the number of rooting strips was 10.81.Tissue culture seedlings could take root outside the bottle.Tissue culture seedlings grew well after transplanting,and the survival rate of transplanting was more than 95.56%.
