摘 要:为了进一步对竹节参组培育苗技术体系进行优化,以竹节参叶片、叶脉、叶柄、茎段、花、花序轴为外植体,进行组培育苗体系优化试验。结果表明,6种竹节参外植体经消毒处理在MS+1.5 mg/L NAA+1.5 mg/L 2,4-D+0.2 mg/L KT配方下均能诱导出愈伤组织,其中愈伤组织诱导率最佳外植体为叶柄和花序轴,诱导率为97%,其次是叶片(95%)、叶脉和茎段(83%)、花(73%)。在优化叶片愈伤组织诱导中发现,3.0 mg/L 2,4-D处理诱导率可达100%,且4级愈伤组织诱导率为30%。叶片在MS+2.0 mg/L NAA+1.5 mg/L 2,4-D+0.6 mg/L KT配方下愈伤组织诱导率为100%,4级愈伤率为23%。叶片愈伤组织在MS+1.5 mg/L NAA+2.5 mg/L 6-BA+1.0 mg/L GA_(3)配方下,培养45 d不定芽诱导率为19%,将愈伤组织在MS+1.5 mg/L NAA+2.4 mg/L 6-BA+1.0 mg/L GA_(3)进行诱导70 d,期间继代1次,不定芽诱导率可达56%,增殖2.95倍。丛生芽在单因素MS+1.0 mg/L NAA诱导下生根诱导率为27%,在多因素组合MS+0.5 mg/L NAA+0.5 mg/L IBA的生根诱导率为53%,相比单因素诱导率提升1.96倍。In order to further optimize the technical system of tissue culture of Panax japonicus,leaf,vein,petiole,stem segment,flower and inflorescence axis of Panax japonicus were used as explants to optimize the system of tissue culture of Panax japonicus.The results showed that 6 explants were treated with disinfection at MS+1.5 mg/L callus could be induced by NAA+1.5 mg/L 2,4-D+0.2 mg/L KT.The best callus induction rate was in petiole and inflorescence axis,reaching 97%,followed by leaves of 95%,leaf veins and stem segments of 83%and flowers of 73%.In the optimization of callus induction,it was found that the induction rate of 3.0 mg/L 2,4-D treatment could reach 100%,and the induction rate of Grade 4 callus was 30%.The callus induction rate of leaves under MS+2.0 mg/L NAA+1.5 mg/L 2,4-D+0.6 mg/L KT formula was 100%,and the Grade 4 callus rate was 23%.The adventist bud induction rate of leaf callus cultured under MS+1.5 mg/L NAA+2.5 mg/L 6-BA+1.0 mg/L GA formula for 45 days was 19%,and callus cultured under MS+1.5 mg/L NAA+2.4 mg/L 6-BA+1.0 mg/L GA after 70 days of induction,the induction rate of indefinite bud was 56%and the proliferation was 2.95 times.The rooting rate of cluster buds under single factor MS+1.0 mg/L NAA induction was 27%,and the rooting rate of multiple factor MS+0.5 mg/L NAA+0.5 mg/L IBA induction was 53%,which was 1.96 times higher than that of single factor induction.
