黑龙江省稻田稗草HL31种群对氰氟草酯的抗性水平及抗性机理分析

摘  要:[目的]为明确稗草Echinochloa crus-galli HL31种群对氰氟草酯的抗性水平及抗性机理。[方法]采用整株生物测定法探究稗草HL31对氰氟草酯抗性水平以及对其他常用除稗剂的多抗性或交互抗性,并研究非靶标酶抑制剂对抗性的影响;采用分子生物学方法测定靶标酶ACCase基因序列和表达量。[结果]稗草种群HL31对氰氟草酯产生高水平抗性,抗性倍数为14.45;对唑酰草胺、敌稗、五氟磺草胺、二氯喹啉酸产生不同程度多抗性或交互抗性,对嘧啶肟草醚敏感;对已报道的7个ACCase CT功能区域位点进行对比,抗性种群HL31和敏感种群HL33未发生氨基酸改变;施药后2者之间靶标酶相对基因表达量达到极显著性差异;P450和GST抑制剂显著降低抗性种群对氰氟草酯的抗性水平。[结论]稗草种群HL31对氰氟草酯产生高水平抗性,可能与靶标酶过量表达、P450及GST介导的非靶标抗性有关,与靶标酶基因位点突变无关。[Aims]The aim is to determine the resistance level and mechanism of Echinochloa crus-galli HL31.[Methods]Whole plant bioassay was used to study the resistance level of E.crus-galli HL31 to cyhalofop-butyl,multi-resistance and cross-resistance to other frequently used paspalum.The influence of non-target enzyme inhibitors on the resistance was also studied.The gene sequence and expression of target enzyme ACCase were determined by molecular biology method.[Results]Population of E.crus-galli HL31 had a high level of resistance to cyhalofop-butyl,with the resistance ratio of 14.45;it had different degrees of multi-resistance and cross-resistance to metamifop,propanil,penoxsulam and quinclorac;it was sensitive to pyribenzoxim.There were no changes in amino acid between resistant population HL31 and sensitive population HL33 among the 7 functional sites of ACCase CT reported.There was a significant difference in the relative gene expression of target enzymes between the two groups after application of cyhalofop-butyl;P450 and GST inhibitors significantly reduced the level of resistance to cyhalofop-butyl.[Conclusions]The high level resistance of HL31 to cyhalofop-butyl may be related to the overexpression of target enzyme,non-target resistance mediated by P450 and GST,but not related to the mutation of target enzyme gene locus.
