摘  要:目的 观察不同引经药配伍大黄-丹参药对肺纤维化大鼠的效应差异,探讨引经药的增效作用。方法 雄性SD大鼠60只,随机分为假手术组、模型组、丹参-大黄药对组(简称药对组)、药对+桔梗组、药对+吴茱萸组和药对+细辛组,除假手术组外,其余各组向气管内缓慢注射5 mg/kg博来霉素建立肺纤维化模型,于造模第2天起,各给药组分别给予相应药物灌胃,连续2 w,末次给药后取血和肺,进行血气分析,苏木素-伊红(HE)和Masson染色观察肺组织病理学,酶联免疫吸附试验(ELISA)测定肺组织中丙二醛(MDA)、超氧化物歧化酶(SOD)、羟脯氨酸(HYP)和还原型谷胱甘肽(GSH)的含量,Western印迹分析肺组织中转化生长因子(TGF)-β1、SMAD同源物(Smad)3、磷酸化(p)-Smad3、α-平滑肌肌动蛋白(SMA)的蛋白表达。结果 与模型组比较,各实验组肺组织病理明显改善,肺系数、肺组织中MDA、HYP含量明显降低,动脉血氧分压(PaO_(2))和肺组织中SOD、GSH水平明显升高,肺组织TGF-β1、p-Smad3和α-SMA表达明显减少(P<0.05)。与药对组相比,药对+桔梗组肺系数和肺组织中MDA、HYP含量明显降低,动脉血PaO_(2)和肺组织中GSH水平明显升高,肺组织TGF-β1、p-Smad3和α-SMA蛋白表达明显下调(P<0.05)。结论 大黄-丹参药对可改善大鼠肺纤维化,其机制可能与抑制TGF-β1/Smad3信号通路有关,桔梗可增强大黄-丹参药对抗肺纤维化作用。Objective To compare the effect difference of herb pair of Rhei Radix et Rhizoma and Salviae Miltiorrhizae Radix et Rhizoma with different medicinal guides in pulmonary fibrosis of rats, and to explore the synergistic effect of medicinal guides.Methods Sixty male SD rats were randomly divided into sham operation group, model group, Salvia Miltiorrhiza Radix herb pair group(herb pair for short), herb pair+Platycodonis Radix group, herb pair+Euodiae Fructus group and herb pair+Asari Radix et Rhizom group. Except sham operation group, the pulmonary fibrosis model was established by slowly injecting 5 mg/kg bleomycin into the trachea. From the second day of the model, each group was given corresponding drugs by intragastric administration for 2 w. Blood and lungs were taken for blood gas analysis after the last administration. The histopathology of lung tissue was observed by hematoxylin-eosin(HE) and Masson staining. The contents of malondialdehyde(MDA), superoxide dismutase(SOD), hydroxyproline(HYP) and reduced glutathione(GSH) in lung tissue were determined by enzyme linked immunosorbent assay(ELISA). The protein expressions of transforming growth factor(TGF)-β1, SMAD homologue(Smad)3, phosphorylated(p)-Smad3 and α-smooth muscle actin(SMA) in lung tissue were analyzed by Western blot.Results Compared with the model group, the pathology of lung tissue in each experimental group was significantly improved, the lung coefficient, the contents of MDA and HYP in lung tissue were significantly decreased, the levels of partial pressure of arterial oxygen(PaO_(2)) in arterial blood, SOD and GSH in lung tissue were increased significantly, and the expressions of TGF-β1, p-Smad3 and α-SMA in lung tissue were significantly decreased(P<0.05). Compared with the herb pair group, the lung coefficient and the contents of MDA and HYP in the lung tissue were significantly decreased, PaO_(2) in arterial blood and the level of GSH in the lung tissue were significantly increased, and the expressions of TGF-β1, p-Smad3 and α-SMA p
