摘 要:目的 采用网络药理学方法探讨艾叶治疗功能性便秘(FC)的作用机制。方法 通过中药系统药理学数据库与分析平台(TCMSP)检索艾叶的相关活性成分,通过GeneCards获取FC的疾病靶点基因,采用Cytoscape软件构建中药-成分-靶点-疾病网络图。通过交互靶点GO和KEGG富集分析研究艾叶和FC的相关生物学过程和信号通路。将艾叶的关键活性成分与核心靶点进行分子对接验证。结果 共筛选出药物靶点301个,活性成分8个,疾病靶点1 842个,药物-疾病共有靶点72个。艾叶治疗FC的核心靶点基因为AKT1、SRC、EGFR、VEGFA、MAPK1。GO功能富集分析得到222个条目,其中细胞组分(CC)47个,生物过程(BP)98个,分子功能(MF)77个。KEGG通路富集分析到120条信号通路,癌症的途径、EGFR酪氨酸激酶抑制剂耐药性、癌症中的蛋白多糖、化学致癌作用-受体激活等通路可能与艾叶防治FC密切相关。分子对接验证表明,部分活性成分与核心靶点具有良好的对接活性。结论 艾叶中的多种活性成分能够作用于多个潜在靶点,并通过多条信号通路发挥治疗FC的作用。Objective To explore the mechanism of Artemisiae argyi in the treatment of functional constipation(FC)by network pharmacology.Methods The active components of Artemisiae argyi were searched by traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP),and the disease target genes of FC were obtained by GeneCards.The network diagram of Chinese medicine-component-target-disease was constructed by Cytoscape software.The related biological processes and signaling pathways of Artemisia argyi and FC were studied by GO and KEGG enrichment analysis of interactive targets.The key active components of Artemisia argyi were verified by molecular docking with the core targets.Results A total of 301 drug targets,8 active components,1842 disease targets and 72 drug-disease common targets were screened out.The core target genes of Artemisia argyi in the treatment of FC were AKT1,SRC,EGFR,VEGFA and MAPK1.GO function enrichment analysis obtained 222 items,including 47 cellular component(CC),98 biological process(BP),and 77 molecular function(MF).KEGG pathway enrichment analysis obtained 120 signaling pathways.Cancer pathways,EGFR tyrosine kinase inhibitor resistance,proteoglycans in cancer,chemical carcinogenic effects-receptor activation and other pathways may be closely related to the prevention and treatment of FC with Artemisia argyi.Molecular docking verification showed that some active ingredients had good docking activity with the core target.Conclusion A variety of active ingredients in Artemisia argyi can act on multiple potential targets and play a role in the treatment of FC through multiple signaling pathways.
