摘 要:目的探讨嗅素蛋白4(OLFM4)在脑出血(ICH)模型大鼠血肿周围组织中的表达情况及对相关凋亡蛋白的影响。方法将雄性SD大鼠随机分为模型组(ICH组)、假手术组(Sham组)、OLFM4下调组(OLFM4siRNA组)、空载质粒组(NC-siRNA组)。ICH组、OLFM4siRNA组、NC-siRNA组均采用尾状核注射凝血酶Ⅶ建立ICH模型;OLFM4siRNA组于术前2周将载有OLFM4siRNA的质粒(5μL)注射于大鼠脑室内,NC-siRNA组于术前2周将不含OLFM4siRNA的空质粒(5μL)注射于脑室,用于排除质粒与手术对结果的干扰。采用Longa评分法筛选出造模成功的大鼠。于术后3 d进行改良神经功能缺损评分(m NSS)后,收集脑组织样本,以HE染色法观察脑组织病变,免疫荧光染色观察OLFM4在脑组织中的表达情况,Western blotting法检测血肿周围脑组织中OLFM4、Caspase-9和B细胞淋巴瘤/白血病-2(BCL-2)表达水平。结果ICH组的m NSS评分及OLFM4、Caspase-9表达水平高于Sham组,BCL-2表达水平低于Sham组(P<0.05);与Sham组相比,ICH组大鼠脑组织破坏严重。OLFM4siRNA组的m NSS评分及Caspase-9表达水平高于ICH组,OLFM4、BCL-2表达水平低于ICH组(P<0.05);与ICH组相比,OLFM4siRNA组大鼠脑组织破坏严重。结论下调OLFM4的表达会加重ICH大鼠神经功能损伤,可能与增加Caspase-9和减少BCL-2表达有关。Objective To investigate the expression of olfactomedin-4(OLFM4)in perihematoma tissues of rats with intracerebral hemorrhage(ICH)model and its effect on related apoptotic proteins.Methods Male SD rats were randomly divided into model group(ICH group),sham operation group(Sham group),OLFM4 down-regulation group(OLFM4siRNA group)and null-loaded plasmid group(NC-siRNA group).ICH model was established by injecting thrombinⅦinto caudate nucleus in the ICH group,the OLFM4siRNA group,the NC-siRNA group;the OLFM4siRNA group was injected with plasmid containing OLFM4siRNA(5μL)into the rat ventricle at 2 weeks before operation,and the NC-siRNA group was injected with empty plasmid without OLFM4siRNA(5μL)into rat ventricle at 2 weeks before operation,which was used to exclude the interference between plasmid and surgery on the results.Rats with successful modeling were screened using the Longa score.The Modified Neurological Severity Score(mNSS)was performed at 3 d after operation,and the brain tissue samples were collected.HE staining was used to observe brain tissue lesions,immunofluorescence staining was used to observe the expression of OLFM4 in brain tissues,and Western blotting was used to detect the expression levels of OLFM4,Caspase-9,and B-cell lymphoma/leukemia-2(BCL-2)in perihematoma brain tissues.Results The mNSS score and the expression levels of OLFM4 and Caspase-9 in the ICH group were higher than those in the Sham group,and the expression level of BCL-2 was lower than that in the Sham group(P<0.05);compared with the Sham group,the brain tissue of the ICH group was severely damaged.The mNSS score and Caspase-9 expression level of the OLFM4siRNA group were higher than those of the ICH group,and the expression levels of OLFM4 and BCL-2 were lower than those of the ICH group(P<0.05);compared with the ICH group,the brain tissue of the OLFM4siRNA group was severely damaged.Conclusion Down-regulation of OLFM4 expression exacerbates neurological impairment in rats with ICH,which may be associated with increased
