摘 要:目的:比较结核分枝杆菌潜伏感染(LTBI)者与健康对照(HC)者蛋白质组学差异表达情况,寻找LTBI新型诊断标志物。方法:采用DIA定量蛋白质组学技术分析两组差异表达蛋白质,并运用生物信息学软件对差异蛋白进行分析。利用酶联免疫吸附试验(ELISA)进行验证,并评估该潜在生物标志物在LTBI诊断中的应用价值。结果:共发现88个蛋白质具有差异性表达,其中32个表达上调(FC>1.5,P<0.05),56个表达下调(FC<0.7,P<0.05)。结合GO、KEGG、COG、互作网络分析,差异表达蛋白(GRK2、PLA2G7和ADAMTSL4)被筛选为LTBI的潜在生物标志物。ELISA结果显示:LTBI组的GRK2、PLA2G7与HC组、其他肺部疾病组比较差异有统计学意义(P<0.05),GRK2和PLA2G7的曲线下面积(AUC)分别为0.716和0.756,灵敏度分别为0.667和0.722,特异度分别为0.917和0.833。结论:本研究将DIA技术与ELISA验证相结合,获得了2种新的LTBI诊断生物标志物(GRK2和PLA2G7),具有较高的灵敏度和特异度,在LTBI的诊断中有较好潜在应用前景。Objective:Comparing the differential expression of proteomes between latent mycobacterium tuberculosis infection(LTBI)and healthy controls(HC),to search for new diagnostic biomarkers for LTBI.Methods:Using DIA quantitative proteomics technology to analyze two groups of differentially expressed proteins,and using bioinformatics software to analyze the differentially expressed proteins.Validate using enzyme-linked immunosorbent assay(ELISA)and evaluate the potential application value of this biomarker in the diagnosis of LTBI.Results:A total of 88 proteins were found to be differentially expressed,of which 32 were up-regulated(FC>1.5,P<0.05)and 56 were down-regulated(FC<0.7,P<0.05).By combining GO,KEGG,COG,and interaction network analysis,differentially expressed proteins(GRK2,PLA2G7,and ADAMTSL4)were screened as potential biomarkers for LTBI.ELISA results showed that GRK2 and PLA2G7 in LTBI group were significantly different from HC group and other lung disease groups(P<0.05).GRK2 and PLA2G7 had area under the curve(AUC)values of 0.716 and 0.756,sensitivity values of 0.667 and 0.722,and specificity values of 0.917 and 0.833,respectively.Conclusion:In this study,DIA technology was combined with ELISA validation to obtain two new diagnostic biomarkers for LTBI(GRK2 and PLA2G7),which have high sensitivity and specificity,and have good potential application prospects in the diagnosis of LTBI.
