摘 要:目的:通过分析hsa-miR-381-3p的保守性及靶基因,探索其可能发挥的生物学作用。方法:首先,通过UCSC基因组浏览器,比较人类与其他物种的基因组序列,分析hsa-miR-381-3p的序列保守性。再利用miR-Gator数据库探索hsa-miR-381-3p在不同器官和疾病中的表达情况。随后,预测hsa-miR-381-3p的潜在靶基因,使用miRDB、mirDIP和TargetScan数据库,并与miRTarBase数据库整合以提高准确性。进行蛋白质—蛋白质相互作用分析,揭示基因相互作用网络。最后,通过DAVID 6.8工具进行GO功能和KEGG通路富集分析,深入理解hsa-miR-381-3p的生物学功能和作用机制。结果:miR-381-3p序列在多个物种间保守,表明其在生物学过程中的重要性和演化稳定性。鉴定出279个与miR-381-3p相互作用的靶基因,包括BRD4、MDM2等,主要功能是与蛋白质和RNA结合,调控转录因子和辅因子活性,在基因表达调控中起关键作用。KEGG通路分析显示,这些靶基因涉及35个信号通路,包括干细胞多能性、轴突导向等,这些通路在细胞生命活动中至关重要。结论:hsa-miR-381-3p在调控关键信号通路中的作用,使其在肿瘤诊断和治疗领域具有应用前景。Objective:To explore the potential biological functions of hsa-miR-381-3p by analyzing its conservation and target genes.Methods:Firstly,we compared the genomic sequences of humans and other species using the UCSC Genome Browser to analyze the sequence conservation of hsa-miR-381-3p.We then utilized the miR-Gator database to investigate the expression of hsa-miR-381-3p in different organs and diseases.Next,we predicted the potential target genes of hsa-miR-381-3p using miRDB,mirDIP,and TargetScan databases,and integrated the results with miRTarBase to improve accuracy.We performed protein-protein interaction analysis to reveal the gene interaction network.Finally,we conducted GO functional and KEGG pathway enrichment analyses using DAVID 6.8 to gain a deeper understanding of the biological functions and mechanisms of hsa-miR-381-3p.Results:The sequence of miR-381-3p was found to be highly conserved across multiple species,indicating its importance and evolutionary stability in biological processes.We identified 279 target genes that interact with hsa-miR-381-3p,including BRD4,MDM2,etc,which are primarily involved in protein-RNA binding,transcription factor regulation,and gene expression regulation.KEGG pathway analysis revealed that these target genes are involved in 35 signaling pathways,including stem cell pluripotency and axon guidance,which are crucial for cellular life activities.Conclusion:The role of hsa-miR-381-3p in regulating key signaling pathways makes it a promising candidate for cancer diagnosis and treatment.
