摘 要:目的 探讨慢性砷中毒大鼠睾丸组织survivin、caspase-3蛋白表达的变化。方法 将40只健康清洁级雄性SD大鼠随机分为4组,分别为对照(蒸馏水)组和2.4、12、60 mg/L亚砷酸钠染毒组,每组10只,采用自由饮水方式连续染毒6个月。采用免疫印迹法测定大鼠睾丸组织内survivin、caspase-3的蛋白表达水平,流式细胞术观察精子的凋亡率,采用改良透明质酸钠-明胶底物膜法检测精子透明质酸酶(HYD)活力。结果 与对照组比较,各浓度亚砷酸钠染毒组大鼠精子凋亡率较高,差异均有统计学意义(P<0.05);且随着亚砷酸钠染毒浓度的升高,大鼠精子凋亡率呈上升趋势。与对照组相比,各浓度亚砷酸钠染毒组大鼠精子透明质酸酶阳性率及反应直径均较低,差异有统计学意义(P<0.05);且随着亚砷酸钠染毒浓度的升高,大鼠精子透明质酸酶阳性率及反应直径呈下降趋势。与对照组相比,各浓度亚砷酸钠染毒组大鼠睾丸组织内survivin蛋白的表达水平均降低,而caspase-3蛋白的表达水平均升高,差异均有统计学意义(P<0.05);且随着亚砷酸钠暴露浓度的升高,大鼠睾丸组织内survivin蛋白的表达水平呈下降趋势,而caspase-3蛋白的表达水平呈上升趋势。大鼠睾丸组织survivin蛋白与caspase-3蛋白表达水平的呈显著负相关(r=-0.878,P<0.01)。结论 砷可能通过诱导survivin表达降低及caspase-3表达升高促进大鼠精子细胞凋亡,从而发挥砷的雄性生殖毒性作用。Objective To understand the changes of survivin and caspase-3 expression in the testis tissue of rats under chronic arsenic exposure.Methods Forty healthy clean level male SD rats were randomly divided into four groups,10 in each group,the control group(distilled water),sodium arsenite exposure groups(2.4,12.0 and 60.0 mg/L respectively).The rats were exposed to sodium arsenite through drinking water for six consecutive months.The expression levels of survivin and caspase-3 in testicular tissue were detected by western blotting,the apoptosis rate of sperm was assessed by using flow cytometry.The activity intensity of HYD was detected by modified sodium hyaluronate-gelatin membrane.Results Compared with the control group,the apoptosis rates of spermatozoa in sodium arsenite exposure groups were significantly higher(P<0.05),with a dose dependent manner.Compared with the control group,the positive rate and reaction diameter of HYD in sodium arsenite exposure groups were significantly lower(P<0.05),with a dose dependent manner.The levels of survivin protein expression were lower and the levels of caspase-3 protein expression were higher in sodium arsenite exposure groups,the differences were statistically significant(P<0.05),with dose dependent manners.The expression of survivin and caspase-3 protein in testicular tissue of rats was negatively correlated(r=-0.878,P<0.01).Conclusion Chronic arsenic exposure can cause apoptosis of spermatogenic cells of rat by down-regulate the expression of survivin and up-regulate the expression caspase-3,thus the toxic effects of arsenic on male reproduction can be exerted.
