摘  要:目的 观察电针对大鼠蛛网膜下腔出血(subarachnoid hemorrhage, SAH)后认知障碍及海马组织Wnt1和β-Catenin蛋白表达的影响,探讨电针对SAH后认知功能障碍的干预效应及其可能机制。方法 清洁级雄性SD大鼠随机分为空白对照组、假手术组、模型组、尼莫地平组、电针组,每组7只。采用经视交叉池注血法制备疾病模型,电针组取“印堂”穴和双“迎香”穴进行电针治疗,尼莫地平组接受鼻饲尼莫地平。应用Morris水迷宫实验对各组大鼠进行行为学检测,评估大鼠认知功能。通过HE染色法观察大鼠海马神经元细胞形态,运用免疫印迹法(Western Blot)技术检测Wnt1、β-Catenin蛋白的表达情况。结果 与空白对照组比较,模型组大鼠学习记忆能力显著下降(P<0.05),海马组织疏松,神经元损伤较严重,细胞排列紊乱,形态结构异常,Wnt1与β-Catenin蛋白表达水平明显降低(P<0.01)。而与模型组比较,电针组大鼠认知功能显著改善(P<0.05),海马区病理改变明显减轻,Wnt1及β-Catenin的表达水平显著增高(P<0.01,P<0.05)。结论 电针疗法能够减缓神经元损伤,对SAH后认知障碍可起良性干预作用,其机制可能与调控Wnt/β-Catenin信号转导通路活性有关。Objective To observe the effect of electroacupuncture on cognitive impairment and expressions of Wnt1 andβ-Catenin in hippocampus after subarachnoid hemorrhage(SAH)in rats and explore the intervention effect of acupuncture on cognitive dysfunction after SAH and its possible mechanism.Methods SD rats were randomly divided into blank control group,sham operation group,model group,nimodipine group and electroacupuncture group,with 7 rats in each group.The disease models were prepared by autologous blood injection into prechiasmatic cistem.Electroacupuncture group had electroacupuncture at Yintang(EX-HN3)and the bilateral Yingxiang(LI20).The nimodipine group had nasal feeding with nimodipine.The Morris water maze experiment was applied to assess cognitive function.The morphology of rat hippocampal neurons was observed by HE staining,and the expressions of Wnt1 andβ-Catenin were detected by Western Blot.Results Compared with the blank control group,the model group significantly reduced learning and memory ability(P<0.05),loose hippocampal tissue,severe neuronal damage,disorganized cell arrangement,abnormal morphological structure,and significantly reduced expression levels of Wnt1 andβ-Catenin protein(P<0.01).Compared with the model group,the rats in the electroacupuncture group significantly improved cognitive function(P<0.05),significantly reduced pathological changes in hippocampus and significantly higher expression levels of Wnt1 andβ-Catenin(P<0.01,P<0.05).Conclusion Electroacupuncture can alleviate neuronal damage and plays a benign inter-vention role in cognitive impairment after SAH.The mechanism may be related to regulating the activity of Wnt/β-Catenin signal transduction pathway.
