摘  要:目的探究快速解冰冻去甘油法在冰冻红细胞解冻中的应用价值。方法选择江苏省宿迁市中心血站保存的30人份7 d内去白细胞的悬浮红细胞,甘油化后于-80℃±5℃中制备成冰冻红细胞产品,保存6个月。将冰冻红细胞产品等量分为两组,每组10袋。对照组采用常规法进行解冻,实验组采用快速解冰冻去甘油法进行解冻。对比两组解冻过程中加溶液混匀时间及解冻后的红细胞指标、质量指标。结果实验组解冻过程中加溶液进行混匀时间短于对照组,P<0.05。两组解冻后的红细胞含量和血细胞比容对比,P>0.05;实验组血红蛋白浓度低于对照组,P<0.05;两组解冻后的红细胞回收率和溶血率对比,P>0.05;实验组游离血红蛋白含量和甘油残留量均低于对照组,P<0.05;实验组ATP含量和2,3-DPG含量均高于对照组,P<0.05。两组均无细菌生成。结论快速解冰冻去甘油法解冻冰冻红细胞快速、安全、有效,可在保证红细胞产品质量的同时,加快红细胞解冻速度,保持红细胞制品的品质稳定,并进一步优化其品质。Objective:To explore the application value of quick freezing and glycerol removal in thawing frozen red blood cells.Methods:30 samples of suspended red blood cells with leukocytes removed within 7 days were selected from Suqian Central Blood Station Jiangsu Province,and then they were glycerol-prepared into frozen red blood cell products at-80℃±5℃and kept for 6 months.The frozen red blood cell products were equally divided into two groups with 10 bags in each group.The control group was thawed by routine method,while the experimental group was thawed by quick unfreezing and glycerol removal method.The mixing time、red blood cell index and quality index after thawing were compared between the two groups.Results:The time of adding solution in the thawing process in the experimental group was shorter than that in the control group,P<0.05.The content of red blood cells and hematocrit after thawing were compared between the two groups,P>0.05;The hemoglobin concentration in the experimental group was lower than that in the control group,P<0.05.The recovery rate and hemolysis rate of red blood cells after thawing were compared between the two groups,P>0.05;The content of free hemoglobin and glycerol residue in the experimental group were lower than those in the control group,P<0.05.The contents of ATP and 2,3-DPG in the experimental group were higher than those in the control group,P<0.05.No bacteria were produced in both groups.Conclusion:The quick unfreezing and glycerol removal method is fast、safe and effective in thawing frozen red blood cells,which can ensure the quality of red blood cell products,accelerate the thawing speed of red blood cells,maintain the quality stability of red blood cell products and further optimize their quality.
