摘  要:目的探讨镉暴露对小鼠精母细胞(GC-2spd细胞)内AMPK/ULK1通路的影响。方法将GC-2spd细胞分别暴露于终浓度为0(对照)、2.5、5、10μmol/L的氯化镉培养液染毒24 h。采用化学荧光法检测细胞活性氧(ROS)的水平,采用Western blot法检测p-AMPKα、p-ULK1、Beclin1和LC3Ⅱ/Ⅰ蛋白的表达水平。结果与对照组相比,2.5、10μmol/L氯化镉暴露GC-2spd细胞内ROS水平较高,差异均有统计学意义(P<0.05)。与对照组相比,5μmol/L氯化镉暴露GC-2spd细胞内p-AMPKα、LC3(Ⅱ/Ⅰ)蛋白的表达水平及10μmol/L氯化镉暴露GC-2spd细胞内p-AMPKα、p-ULK1、Beclin1和LC3Ⅱ/Ⅰ蛋白的表达水平均较高,差异有统计学意义(P<0.05);且随着氯化镉暴露浓度的升高,GC-2spd细胞内p-AMPKα、p-ULK1、Beclin1和LC3Ⅱ/Ⅰ蛋白的表达水平均呈上升趋势。结论镉可能促使GC-2spd细胞内ROS生成并激活AMPK/ULK1信号通路诱导小鼠精母细胞发生自噬。Objective To investigate the effect of cadmium exposure on AMPK/ULK1 pathway in mouse spermatocytes(GC-2spd).Methods GC-2spd cells were exposed to cadmium chloride at the doses of 0(control),2.5,5,and 10μmol/L respectively,for 24 hours.The level of reactive oxygen species(ROS)in the cells was detected by using a chemical fluorescence method,and the expression levels of p-AMPKα,p-ULK1,Beclin1,and LC3Ⅱ/Ⅰproteins were measured by Western blot.Results Compared with the control group,exposure to 2.5 and 10μmol/L cadmium chloride resulted in significantly higher levels of ROS in GC-2spd cells(P<0.05).Compared with the control group,exposure to 5μmol/L cadmium chloride resulted in higher expression levels of p-AMPKαand LC3(Ⅱ/Ⅰ)proteins in GC-2spd cells,while exposure to 10μmol/L cadmium chloride resulted in higher expression levels of p-AMPKα,p-ULK1,Beclin1,and LC3Ⅱ/Ⅰproteins in GC-2spd cells,with significant differences(P<0.05).Furthermore,as the concentration of cadmium chloride exposure increased,the expression levels of p-AMPKα,p-ULK1,Beclin1,and LC3Ⅱ/Ⅰproteins in GC-2spd cells showed an upward trend.Conclusion Cadmium may induce the generation of ROS and activate the AMPK/ULK1 signaling pathway,leading to autophagy in mouse germ cells(GC-2spd cells).
