摘 要:肝细胞是肝脏组织的主要组成部分,牛的生产、繁殖、健康和饲料转化效率均与肝脏密切相关,对牛肝细胞的代谢和解毒功能进行研究具有重要意义。但肝细胞存活时间短、特异性功能活性逐渐丧失,缺少成熟的细胞系用于牛肝细胞特异性功能研究。文章系统阐述了牛肝细胞的体外培养方法,肝细胞的去分化机制、相关信号途径及应对肝细胞去分化的策略,分离过程肝细胞的凋亡机制,以及肝细胞的冷冻保存方法,以期建立便捷高效的肝细胞体外培养体系,为获得存活时间长、能稳定表达特异性活性的肝细胞提供理论基础。Hepatocytes are the primary cells in liver tissue,which are closely linked to cattle production,reproduction,health and feed conversion efficiency.Understanding the metabolism and detoxification functions of bovine hepatocytes is of great significance.However,due to their short lifespan and the progressive loss of functional activity,there are currently no well-established cell lines for bovine hepatocyte-specific studies.This review provides a comprehensive overview of in vitro culture methods for bovine hepatocytes,addressing key challenges such as hepatocyte dedifferentiation and the associated signaling pathways.Additionally,it explores strategies to counteract dedifferentiation and reveals the mechanisms of hepatocyte apoptosis during isolation.Techniques for cryopreservation were also discussed herein,with the aim of developing a practical and efficient in vitro hepatocyte culture system.Ultimately,this research seeks to offer a foundation for obtaining hepatocytes with extended survival times and stable functional activity,offering valuable insights for future studies.
