摘 要:目的研究青麦饮对实验性干燥综合征(experimental Sjogrens syndrome,ESS)小鼠干预后疾病的发展变化及B细胞亚群分布比例的影响。方法将24只C57BL/6小鼠随机分为正常组、模型组、青麦饮低剂量组(1.125 g/mL)、青麦饮高剂量组(2.25 g/mL),每组6只。通过对小鼠皮下注射唾液腺蛋白乳剂构建ESS小鼠模型,造模成功后连续灌胃6周。灌胃结束后进行唾液流率检测,ELISA法检测血清抗SSA抗体和抗M3R抗体水平。观察小鼠脾脏及颈部淋巴结大小形态的变化,制备各组小鼠脾脏、颈部淋巴结、骨髓单细胞悬液,流式细胞术检测各B细胞亚群分布比例。结果与正常组比较,模型组小鼠唾液流率下降(P<0.01);脾脏和颈部淋巴结均有明显增大(P<0.01);抗SSA抗体、抗M3R抗体水平升高(P<0.05);脾脏和颈部淋巴结生发中心B细胞比例均升高(P<0.05),脾脏和骨髓中的浆细胞比例均有升高(P<0.05),脾脏边缘区B细胞比例下降(P<0.05),滤泡B细胞比例上升(P<0.01)。与模型组比较,青麦饮干预组唾液流率增加(P<0.05);抗M3R抗体水平下降,高剂量组更为显著(P<0.05)。青麦饮高剂量组抗SSA抗体水平显著降低(P<0.01);脾脏和颈部淋巴结均有不同程度的减小(P<0.05),脾脏边缘区B细胞比例上升(P<0.01),滤泡B细胞比例下降(P<0.05)。结论青麦饮可能通过调控B细胞亚群减轻ESS干燥症状,改善抗体水平,缓解ESS小鼠的疾病进程。Objective To study the effect of Qingmai Yin on the treatment and B cell subpopulations in mice with experimental Sjogren's syndrome(ESS).Methods Twenty-four C57BL/6 mice were randomly divided into normal group,model group,low-dose group of Qingmai Yin(1.125 g/mL),and high-dose group of Qingmai Yin(2.25 g/mL),six mice in each group.The ESS mouse model was constructed by subcutaneous injection of salivary gland protein emulsion into the mice,and after successful modelling,the mice were gavaged continuously for 6 weeks.At the end of gavage,salivary flow was detected,and serum anti-SSA antibody and anti-M3R antibody levels were detected by ELISA.Changes in the size and morphology of the spleen and cervical lymph nodes of mice were observed,and single-cell suspensions of the spleen,cervical lymph nodes,and bone marrow of mice in each group were prepared,and the proportion of distribution of each B-cell subpopulation was detected by flow assay.Results Compared with the normal group,the saliva flow in the model group decreased(P<0.01),the spleen and cervical lymph nodes were significantly increased(P<0.01).The level of ant-ssa and anti-M3R antibody increased(P<0.05),the proportion of GCB cells in spleen and cervical lymph nodes increased(P<0.05),the proportion of plasma cells in spleen and bone marrow increased(P<0.05),the proportion of B cells in the marginal area of the spleen decreased(P<0.05),and the proportion of follicular B cells increased(P<0.01)。Compared with the model group,the saliva flow in the Qingmai Yin intervention group increased(P<0.05),and the level of anti-M3R antibody decreased in the high-dose group(P<0.05).The level of anti-SSA antibody in the high-dose group was significantly decreased(P<0.01),the lymph nodes in the spleen and neck decreased to varying degrees(P<0.05),the proportion of B cells in the marginal zone of the spleen increased(P<0.01),and the proportion of follicular B cell subsets decreased(P<0.05)Conclusion Qingmai Yin may alleviate the dry symptoms of ESS,improve the antibody lev
