摘 要:目的 探究健脾止动汤对慢性束缚应激抽动障碍(TD)大鼠多巴胺(DA)系统的影响。方法 选择48只雄性SD大鼠,随机分为空白组、模型组、硫必利组与健脾止动汤组,每组12只。干预结束后,高效液相色谱(HPLC)法检测脑组织中的DA、二羟基苯乙酸(DOPAC)和高香草酸(HVA)含量;Western Blot和RT-PCR检测脑中多巴胺D1受体(DRD1)、DRD2蛋白及mRNA表达。结果 与空白组比较,模型组大鼠海马组织中DA、HVA含量,下丘脑组织中DA、DOPAC含量、DRD2蛋白、DRD1 mRNA、DRD2 mRNA表达明显升高(P<0.05);与模型组比较,硫必利组下丘脑DA、DOPAC含量、DRD2蛋白表达明显降低,健脾止动汤组海马组织中DA、DOPAC、HVA含量,下丘脑组织中DA、DOPAC、HVA含量、DRD1蛋白、DRD1 mRNA表达明显降低(P<0.05);与硫必利组比较,健脾止动汤组海马组织中HVA含量明显降低(P<0.05)。结论 健脾止动汤干预能够通过DRD1相关靶点降低慢性束缚应激抽动障碍大鼠海马和下丘脑组织中DA、DOPAC、HVA含量,改善多巴胺系统功能,达到抗抽动作用。Objective To investigate the effect of Jianpi Zhidong Decoction on dopamine(DA)system in rats with chronic restraint stress tic disorder(TD).Methods 48 male SD rats were selected and randomly divided into the control group,the model group,the tiapride group and the Jianpi Zhidong Decoction group,with 12 rats in each group.After the intervention,the levels of DA,dihydroxyphenylacetic acid(DOPAC)and homovanillic acid(HVA)in brain tissue were detected by high-performance liquid chromatography(HPLC);Western Blot and RT-PCR were used to detect the levels of dopamine D1 receptor(DRD1)and DRD2 proteins,and mRNA expression in the brain.Results Compared with the blank group,the levels of DA and HVA in the hippocampus,the levels of DA,DOPAC and DRD2 protein,and DRD1 mRNA and DRD2 mRNA expression in the hypothalamus in the model group were significantly increased(P<0.05);Compared with the model group,the levels of DA and DOPAC,and the expression of DRD2 protein in the hypothalamus in the tiapride group were significantly decreased(P<0.05);The levels of DA,DOPAC and HVA in hippocampal tissue,the levels of DA,DOPAC and HVA,and DRD1 protein and DRD1 mRNA expression in hypothalamic tissue were significantly decreased in the Jianpi Zhidong Decoction(P<0.05);Compared with the tiapride group,the HVA content in the hippocampus in the Jianpi Zhidong Decoction group was significantly decreased(P<0.05).Conclusion Jianpi Zhidong Decoction intervention can reduce the levels of DA,DOPAC and HVA in the hippocampus and hypothalamus of TD rats with chronic restraint stress Tic Disorders through DRD1-related targets,and improve the function of the DA system,thereby achieving anti-tic effect.
