摘 要:利用HPLC检测方法,研究金丝桃素在不同的超声时间、微波功率、温度、溶剂、中长期稳定性情况,为高效制备及后续应用研究提供参考。HPLC法能获得基线稳定、分离度高的金丝桃素图谱;检测限(LOD)为5.2 mg/L、定量限(LOQ)为10.4 mg/L;在浓度为10.4~124.8 mg/L的线性范围内线性良好,线性方程y=34 774.7x-349 830.4(R^(2)为0.999 27)。金丝桃素稳定性研究结果显示,在超声处理2 h内稳定性良好;温敏性测试表明-20℃为最佳保存温度,但在常温或低于60℃时,可短期避光存储;微波功率550 W时稳定性最佳;10 d内的中长期稳定性实验表明,高温、低温和强光都导致金丝桃素溶液不稳定。To study the stability of hypericin at different ultrasonic time,microwave power,temperature,solvent and medium and long term by HPLC,and to provide reference for efficient preparation and subsequent application.The chromatogram of hypericin with stable baseline and high separation was obtained by HPLC.The limit of detection(LOD)was 5.2 mg/L,and the limit of quantitation(LOQ)was 10.4 mg/L.In the linear range from 10.4 mg/L to 124.8 mg/L,the linear equation y=34774.7x-349830.4(R^(2)=0.99927)is well linear.The results of hypericin stability study showed that the stability of hypericin was good within 2 h after ultrasonic treatment.Temperature sensitivity test shows that-20℃is the best storage temperature,but at room temperature or below 60℃,it can be stored away from light for a short time;the stability is best when the microwave power is 550 W.The medium and long term stability experiments within 10 d showed that high temperature,low temperature and strong light all caused the instability of hypericin solution.
