摘　　要：猪流行性腹泻(PED)是猪流行性腹泻病毒(PEDV)引起的一种具有高度传染性的仔猪肠道疾病。本研究利用HEK-293F细胞真核表达系统表达了PEDV S基因不同截短蛋白(S、S1和CTD蛋白),免疫4周龄的BALB/c小鼠,分别于免疫后0 d、14 d和28 d采集小鼠血清。通过ELISA和病毒中和试验证实3种蛋白均能使小鼠产生良好的免疫应答效果;进一步采用流式细胞术检测CD3^(+)CD4^(+)CD8^(+)淋巴细胞亚型比例以及小鼠细胞IFN-γ、IL-4因子检测来评估PEDV亚单位疫苗免疫抗体效价,结果表明其中以PEDV S蛋白免疫原性最好。本研究为PED亚单位疫苗的研发提供了理论支持。Porcine epidemic diarrhea(PED)is a highly contagious intestinal disease of piglets,which is caused by porcine epidemic diarrhea virus(PEDV).In this study,we used the HEK-293F cells to express different truncated proteins of the PEDV S gene(S,S1,and CTD).Four-week-old BALB/c mice were immunized with these proteins and serum samples were collected at 0,14 and 28 d post immunization.These three proteins were able to induce favorable immune responses in mice as confirmed by ELISA and virus neutralization.The proportion of CD3^(+)CD4^(+)CD8^(+)lymphocyte subsets was determined in flow cytometry and IFN-γin mouse cells and IL-4 were also examined to assess the immunogenicity of these truncated proteins.The results indicated that the PEDV S protein was the most immunogenic among these truncated proteins.This study provided helpful information for the development of a PEDV subunit vaccine.
