摘  要:为了解规模化猪场副猪嗜血杆菌(HPS)的耐药基因流行情况,对来自国内52株HPS分离株开展了耐药基因检测、药敏试验、氨基糖苷类耐药基因Aph(3')-Ia和StrB氨基酸序列的比对及分析。结果表明:HPS分离株含有多重耐药基因(β-内酰胺类,氨基糖苷类,磺胺类药和Ⅰ类整合子),其中bla-_(TEM)、Aph(3')-Ia、3'CS和Int1基因检出率为100%;HPS分离菌株对硫酸链霉素的耐药率高达75%。HPS分离菌株Aph(3')-Ia编码的部分氨基酸与除了已知HPS之外的6种细菌的氨基酸序列相同;HPS分离株JX-6 StrB基因编码的氨基酸序列与其他细菌差别较大,该菌株第18号位点StrB基因的ATP结合位点也发生了突变。本研究表明规模化养猪场HPS氨基糖苷类耐药基因Aph(3')-Ia和StrB携带率高,细菌耐药性的差异和耐药基因的改变可能是由一些特定的位点突变所导致。本研究为规模化养殖场HPS耐药基因变化规律研究和相关疾病的防控奠定了基础。This study was aimed to investigate the drug resistance and responsible genes of Haemophilus parasuis(HPS)on large-scale pig farms in China.The drug resistance phenotypes and genes of 52 HPS isolates were examined and amino acid sequences of aminoglycoside resistance genes Aph(3')-Ia and StrB were also compared and analyzed in the present study.The results showed that multi-drug resistance genes were detected in these isolates,such asβ-lactams,aminoglycosides,sulfa drugs and class I integrons.The presence of resistance genes bla-_(TEM),Aph(3')-Ia,3'CS and Int1 were detected in 100%isolates.The resistance to streptomycin sulfate was found in 75%isolates.According to the sequence analysis of Aph(3')-Ia,some amino acids were identical to those of 6 other bacterial species rather than the known HPS.The amino acid sequence of StrB gene of the HPS strain JX-6 was significantly different from those of other bacteria at sites 14-19.Meanwhile,the site 18 of JX-6 StrB gene for the ATP binding was found mutated.Our study indicated a high prevalence of resistance genes Aph(3')-Ia and StrB of aminoglycoside drugs in HPS on large-scale pig farms and the differences in bacterial drug resistance and changes of resistance genes might be caused by mutation of some specific sites.This study laid a foundation for the study of the changes of HPS drug resistance genes on large-scale breeding farms and gave valuable insight into the prevention and control of related diseases.
