基于神经细胞凋亡相关因子Caspase-3,NMDAR探讨当归芍药散对血管性痴呆的影响

摘  要:目的探讨当归芍药散对血管性痴呆大鼠海马组织、神经凋亡相关因子Caspase-3,NMDAR的影响。方法大鼠随机分为假手术组,模型组,阳性对照组,当归芍药散低剂量组(DSS低剂量组),当归芍药散高剂量组(DSS高剂量组),除假手术组外均采用双侧颈总动脉永久结扎法制备血管性痴呆大鼠模型,术后根据组别分别采用蒸馏水、尼莫地平、当归芍药散灌胃,持续4周,采用Morrirs水迷宫实验检测血管性痴呆大鼠记忆能力,采用Western blot法检测海马组织中Caspase-3,NMDAR蛋白的表达。结果与假手术组比较,模型组的水迷宫实验结果均显著低于假手术组(P<0.05),海马组织中的Caspase-3、NMDAR蛋白质含量显著升高(P<0.05)。与模型组比较,阳性对照组、当归芍药散高剂量组与当归芍药散低剂量组的水迷宫实验结果明显改善(P<0.05),阳性对照组和当归芍药散高剂量组的Caspase-3,NMDAR蛋白质含量均显著降低(P<0.05)。结论当归芍药散通过减少神经细胞凋亡发挥对血管性痴呆的治疗作用。Objective To explore the effects of Danggui Shaoyao San(DSS)on hippocampal tissue and apoptosis related factors Caspase-3 and NMDAR in rats with vascular dementia.Methods The rats were randomly divided into a sham operation group,a model group,a positive control group,a low-dose group of Danggui Shaoyao San(DSS low-dose group),and a high-dose group of Danggui Shaoyao San(DSS high-dose group).Except for the sham operation group,a vascular dementia rat model was prepared by permanent ligation of bilateral common carotid arteries.After surgery,distilled water,nimodipine,and Danggui Shaoyao San were orally administered to each group once a day for 4 weeks.The memory and learning capacity of VD rats were tested by the Morris water maze test,and the protein expression of Caspase-3 and NMDAR in hippocampal tissue was detected by Western blot.Results Compared with the sham operation group,the water maze test results were significantly lower in the model group(P<0.05),and the protein contents of Caspase-3 and NMDAR in the hippocampus were significantly increased(P<0.05).Compared with the model group,the water maze test results of the positive control group,DSS high-dose group,and DSS low-dose group were significantly improved(P<0.05).The protein contents of Caspase-3 and NMDAR in the positive control group and DSS high-dose group were significantly reduced(P<0.05).Conclusion Danggui Shaoyao San has a therapeutic effect in the treatment of vascular dementia by reducing neuronal apoptosis.
