摘 要:采用液相色谱-质谱联用非靶向代谢组学方法,分析了配合饲料和饵料鱼两种饲喂方式下翘嘴鳜(Siniperca chuatsi)肠内容物代谢谱的差异,通过主成分分析与数据库比对,筛选差异代谢物及代谢通路,为鳜鱼养殖及其生理代谢研究提供参考。结果显示:通过代谢组学分析,两组间肠道代谢谱存在显著差异,在正离子和负离子模式下共筛选出735种差异代谢物,其中43种代谢物在饲料鳜组显著上调,7种代谢物在饲料鳜组显著下调;KEGG通路富集分析表明差异代谢物主要富集在ABC转运、组氨酸代谢、精氨酸-脯氨酸代谢、胆汁分泌、嘧啶代谢、核苷酸代谢等通路中。研究结果初步表明,配合饲料显著改变了鳜的肠道代谢特征。Siniperca chuatsi is a typical carnivorous fish,of which compound feed has been successfully used to substitute forage fish in its intensive farming.However,the difference in its intestinal metabolism under different diets is still unknown.A metabolomics study was performed on the gut content of S.chuatsi fed with compound feed and forage fish to investigate the dietary effects on the intestinal metabolism of S.chuatsi.A non-targeted metabolomic method using liquid chromatography-mass spectrometry(LC-MS)was applied to investigate the gut metabolic profiles of S.chuatsi,thereafter,principle component analysis and database comparison were used to screen differential metabolites and metabolic pathways.The results showed significant differences in gut content metabolite profiles between the compound feed group and the forage fish group.A total of 735 different metabolites were screened under positive and negative ion modes,among which 43 were significantly up-regulated in the compound feed group and 7 were significantly down-regulated in the compound feed group.KEGG pathway enrichment analysis showed that differential metabolites were mainly concentrated in ABC transport,histidine metabolism,arginine-proline metabolism,bile secretion,pyrimidine metabolism,nucleotide metabolism and other pathways.Results showed that the compound feed significantly changed the intestinal metabolic characteristics of S.chuatsi.The results of this study can provide scientific reference for the intestinal metabolism of S.chuatsi and its adaptation mechanism to compound feed.
