摘 要:为了测定产地湖北恩施黄连醇提液的生物碱含量,研究其抑菌机制,用高效液相色谱法对湖北恩施5年生黄连醇提液主要成分进行测定,并测定了黄连醇提液对大肠埃希氏菌、鸡白痢沙门氏菌、禽多杀性巴氏杆菌和金黄色葡萄球菌的最小抑菌浓度、生长特性、胞外碱性磷酸酶含量、呼吸代谢相关酶类活性的影响。结果显示,醇提液主要成分为小檗碱、表小檗碱、巴马汀和黄连碱,含量分别为7.48%±0.27%、3.52%±0.12%、2.21%±0.07%、2.48%±0.08%,且黄连醇提液对大肠埃希氏菌、鸡白痢沙门氏菌、金黄色葡萄球菌、禽多杀性巴氏杆菌的MIC值分别为0.125、0.5、0.5、0.125 mg/mL。MIC浓度的黄连醇提液可以抑制细菌生长。黄连醇提液还可增加胞外碱性磷酸酶的含量,降低琥珀酸脱氢酶、苹果酸脱氢酶活性。湖北恩施5年生黄连醇提液含量均高于药典规定含量,且具有广谱抑菌活性,可通过破坏细菌膜的通透性、抑制呼吸系统代谢等发挥抑菌效应。This study aimed to determine the components and antibacterial mechanism of the alcohol extract of Coptis chinensis from Enshi,Hubei province.High-performance liquid chromatography(HPLC)was used to determine the main components of the alcohol extract of 5-year-old Coptis chinensis and the minimum inhibitory concentrations(MIC)of the alcohol extract of Coptis chinensis against Escherichia coli,Salmonella pullorum,Pasteurella multocida and Staphylococcus aureus were measured.In addition,the effects of alcohol extract of Coptis chinensis on the growth characteristics,extracellular alkaline phosphatase contents,and enzyme activities related to respiratory metabolism of these bacteria were measured.The result of HPLC showed that the main components of the alcohol extract of Coptis chinensis were berberine,epiberberine,palmatine and coptisine with content levels of 7.48%±0.27%,3.52%±0.12%,2.21%±0.07%,2.48%±0.08%,respectively.The MICs against E.coli,S.pullorum,S.aureus and P.multocida were 0.125,0.5,0.5,0.125 mg/mL,respectively.The growth of bacteria was inhibited by the alcohol extract of Coptis chinensis in MICs.In addition,the alcohol extract of Coptis chinensis increased the extracellular content of alkaline phosphatase and decreased the activities of succinate dehydrogenase and malate dehydrogenase.Furthermore,the alcohol extract of 5-year-old Coptis chinensis from Enshi,Hubei has higher content than that of the standard reported in the Chinese Pharmacopoeia.The alcohol extract of Coptis chinensis also had broad-spectrum antibacterial activity and exerted its antibacterial effects by disrupting the permeability of bacterial membrane and inhibiting respiratory metabolism.
