摘 要:SMYD 4基因编码含有SET结构域和MYND结构域4的赖氨酸甲基转移酶,参与基因表达与个体发育的调控。家蚕中肠组织表达谱分析发现,家蚕核型多角体病毒(BmNPV)抗性品系AN与易感品系C108之间BmSMYD4-X1基因表达存在显著差异。采用分子克隆技术克隆了BmSMYD4-X1基因,通过构建表达载体pIZT/V5-His-mCherry-BmSMYD4-X1转染BmN细胞,发现细胞中BmSMYD4-X1基因过表达能明显抑制BmNPV的基因组复制及病毒增殖,BmNPV的lef-3、vp 39和p 10基因的表达也受到明显抑制。通过RNA干扰能下调BmN细胞内BmSMYD4-X1表达水平约70%,但对BmNPV病毒增殖的影响不明显。上述结果表明,宿主细胞内BmSMYD4-X1基因的表达上调是家蚕对BmNPV抵抗性的机制之一,BmSMYD4-X1可以作为家蚕抗BmNPV分子育种的新基因资源。SMYD 4 is a gene encoding lysine methyltransferase containing SET and MYND domain 4,which is involved in gene expression and developmental regulation.The expression profiling analysis of silkworm midgut tissue showed that there were significant differences in BmSMYD4-X1 gene expression between Bombyx mori nucleopolyhedrovirus(Bm-NPV)resistant strain AN and susceptible strain C108.BmSMYD4-X1 gene was cloned by molecular cloning technology,and BmN cells were transfected with the constructed expression vector pIZT/V5-His-mCherry-BmSMYD 4-X1.It was found that the overexpression of BmSMYD4-X1 gene in BmN cell could significantly inhibit the genome replication of BmNPV and viral proliferation,and the expression of lef-3,vp 39 and p 10 genes of BmNPV were also significantly inhibited.BmSMYD4-X1 expression in BmN cell could be downregulated about 70%through RNA interference,but the effect on BmNPV proliferation was not significant.The above results showed that the upregulated expression of BmSMYD4-X 1 gene in host cells is one of the mechanisms of silkworm resistance to BmNPV,and BmSMYD4-X1 can be used as a new gene resource for the breeding of silkworms resistant to BmNPV.
