摘  要:目的:发热伴血小板减少综合征病毒(severe fever with thrombocytopenia syndrome virus,SFTSV)是一种新发的蜱传病原体。探讨小鼠肠道菌群在SFTSV传播中的作用。方法:通过抗生素治疗和微生物再定殖对昆明小鼠肠道菌群进行调控。然后将感染SFTSV的长角血蜱成蜱和未感染的若蜱共同吸血小鼠。采用实时荧光定量PCR(qPCR)方法检测饱血若蜱体内SFTSV水平。结果:将粪便均匀涂抹在LB琼脂平板上,证实肠道细菌的改变。与对照组和再定殖组相比,抗生素治疗组的微生物群落多样性较单一。在共吸血传播模型中,qPCR分析显示抗生素治疗组的病毒载量较对照组和再定殖组有所增加。结论:小鼠肠道菌群可抑制SFTSV的传播。研究结果为进一步探索SFTSV传播中宿主肠道功能菌提供了基础。Objective:To investigate the role of mouse gut microbiota in the transmission of severe fever with thrombocytopenia syndrome virus(SFTSV),a newly emerging tick-borne pathogen.Methods:The gut microbiota of Kunming mice was manipulated by antibiotics treatment and microbial recolonization.Then,infected adults and uninfected nymphs were co-fed on the mice.The SFTSV levels were determined in the engorged nymphs using quantitative real-time PCR(qPCR)analysis.Results:The alteration of gut bacteria was confirmed by plating the homogenate of faeces onto Luria-Bcrtani(LB)agar plates.Microbial community displayed limited variety in the antibiotic treatment group compared to the control group and the recolonization group.In the model of co-feeding transmission,qPCR analysis revealed that the viral load increased in the antibiotic treatment group compared to the control group and the recolonization group.Conclusion:Mouse gut microbiota can inhibit the transmission of SFTSV.The results provide a basis for further exploration of functional bacteria of host gut in SFTSV transmission.
