摘　　要：为探究葛根素(puerarin,Pur)对湖羊精液冷冻保存效果的影响,在湖羊精液冷冻稀释液中添加不同浓度[0(对照组)、25、50、75和100μmol·L^(-1)]的Pur进行冷冻保存试验,通过检测精子活率、活力、直线速率、曲线速率、平均路径速率、鞭打频率、质膜完整率和顶体完整率等指标,筛选湖羊精液冷冻保存稀释液中Pur的最优添加浓度。结果表明:75μmol·L^(-1)Pur组湖羊精液解冻后精子活率、活力、直线速率、曲线速率、顶体完整率和质膜完整率较对照组显著提高,多数指标也显著高于25、50μmol·L^(-1)Pur组;与75μmol·L^(-1)Pur组相比,100μmol·L^(-1)Pur组多数指标显著下降。综上,在冷冻稀释液中添加不同浓度的Pur均可提高湖羊冷冻精液品质,最适添加浓度为75μmol·L^(-1)。In order to investigate the effect of puerarin(Pur)on the cryopreservation of Hu sheep semen,various concentrations[0(control),25,50,75,and 100μmol·L^(-1)]of Pur were added to the frozen dilution of Hu sheep semen for the cryopreservation test.The optimal concentration of Pur in the cryopreserved dilution was determined by assessing sperm motility,motility,linear rate(VSL),curve velocity(VCL),average path rate,whipping frequency,plasma membrane integrity rate,and acrosome integrity rate.The results indicated that compared to the control group,the sperm motility,motility,linear rate,curve rate,acrosome integrity rate,and plasma membrane integrity rate in the semen thawing group with 75μmol·L^(-1)Pur were significantly higher than those in the control group.Most of these parameters were also significantly elevated compared to the 25 and 50μmol·L^(-1)Pur groups.However,in the 100μmol·L^(-1)Pur group,most of the parameters showed a significant decrease compared to the 75μmol·L^(-1)Pur group.In conclusion,the addition of different concentrations of Pur to the frozen diluent can enhance the quality of frozen semen in Huyang sheep,with the optimal concentration being 75μmol·L^(-1).
