外源ACC处理对翠冠梨果锈及表皮蜡质的影响

摘  要:植物果实表皮蜡质含量和组分变化与果锈形成密切相关,蜡质沉积由1-氨基环丙烷-1-羧酸(1-aminocyclopropane-1-carboxylic acid,ACC)调节,但果锈和蜡质组成如何受ACC影响尚不明确。选用翠冠梨果实为试材,用ACC(100μmol·L^(-1))进行浸没处理,研究其对果锈发生及表皮蜡质含量、组分、相关合成基因表达的影响。结果表明:外源ACC处理可有效减轻梨果锈的发生;较之对照(未进行任何处理),外源ACC使成熟果实果皮角质层增厚24.8%、蜡质总量和烷烃组分含量分别增加29%、122%,其中二十七烷(C27)、三十烷(C30)增加幅度最大;ACC也使蜡质相关基因PpyKCS15、PpyKCS19、PpyKCS24、PpyLTP36、PpyLTP39等相对表达上调,从而促进梨果皮蜡质的生物合成。这说明外源ACC很可能是通过促进梨果实表皮蜡质合成而减缓果锈发生。Dynamic changes in cuticular waxes are strongly associated with fruit russeting,and their deposition are regulated by 1-aminocyclopropane-1-carboxylic acid(ACC).However,the effect of ACC on cuticular wax deposition and skin russeting of Cuiguan pear fruit remains unclear.In this study,Cuiguan pear fruits were immmersed with 100μmol·L^(-1)ACC during fruit development,and its effect on fruit skin russeting,cuticular wax content,composition and the expression patterns of the genes related to cuticular wax synthesis were studied.The results showed that exogenous ACC treatment could effectively inhibit the occurrence of fruit russeting.Compared to the control(untreated)fruits,exogenous ACC treatment thickened the mature fruit skin's cuticle by 24.8%,the contents of total wax and alkanes substances of the mature ACC-treated fruits increased by 29%and 122%respectively,and the contents of heptacosane(C27 alkane)and triacontane(C30 alkane)exhibited the most significant increase.Expression profile analysis revealed that the transcript abundance of PpyKCS15,PpyKCS19,PpyKCS24,PpyLTP36 and PpyLTP39 were significantly higher in ACC-treated fruits and were consistent with the accumulation of cuticular waxes,which might be the key genes involved in ACC-promoted cuticular wax synthesis.These results suggest that ACC treatment promotes cuticular wax synthesis and inhibit the occurrence of skin russeting of Cuiguan pear fruit.
