两种栽培介质条件下根区加压对平邑甜茶根系构型和Mh-PIEZO1/2表达的影响

摘  要:在草炭栽培和水培条件下,对平邑甜茶(Malus hupehensis var.pinyiensis)根系进行加压处理,分析根系构型及MhPIEZO1/2基因表达的变化。结果表明,在草炭栽培条件下,外加20~80 kPa压力显著降低根系长度、根尖数、根系连接数和根系分叉数,20 kPa压力显著增加侧根数量,80 kPa压力显著降低侧根数量;在水培条件下,外加0.5~3.0 kPa压力显著抑制根系生长,根尖数、根系连接数和根系分叉数也显著降低;0.5 kPa显著增加侧根数,3.0 kPa压力显著降低侧根数。平邑甜茶MhPIEZO1和MhPIEZO2编码蛋白含有典型的PIEZO特征结构域;MhPIEZO1在根系中表达量最高,MhPIEZO2在花和芽中有较高表达水平。不论在草炭栽培还是水培条件下,MhPIEZO1/2在根中的表达量均随着外加压力的增加而显著升高,与根系长度、根尖数、根系连接数和根系分叉数对外加压力的反应相反。总体表明,平邑甜茶根系发育和MhPIEZO表达在水培条件下比在草炭栽培条件下更易受到外加压力的影响,根系MhPIEZO表达可能与根系生长存在一定关系。Under peat cultivation and hydroponics,the root system of Malus hupehensis var.pinyiensis was subjected to pressure treatment to analyze the root morphology and expression changes of MhPIEZO1/2.The results showed that under the conditions of peat cultivation,applying a pressure of 20-80 kPa significantly reduced root length,root tip number,root connection number,and root branching number,20 kPa pressure significantly increased the number of lateral roots,and 80 kPa pressure significantly reduced the number of lateral roots.Under hydroponic conditions,applying a pressure of 0.5-3.0 kPa significantly inhibited root growth,and the number of root tips,root connections number,and root branching number also decreased significantly,0.5 kPa pressure significantly increased the number of lateral roots,and 3.0 kPa pressure significantly reduced the number of lateral roots.The proteins encoded by MhPIEZO1 and MhPIEZO2 in M.hupehensis contain typical PIEZO characteristic domains.MhPIEZO1 has the highest expression level in roots,while MhPIEZO2 has a higher expression level in flowers and buds.Under both peat cultivation and hydroponics,the expression level of MhPIEZO1/2 significantly increased with the increase of external pressure,which was opposite to the response of root length,root tip number,root connection number,and root branching number to external pressure.Overall,the root development and MhPIEZO expression of M.hupehensis are more susceptible to external pressure under hydroponics than under peat cultivation,the expression of MhPIEZO in roots may be related to root growth.
