摘  要:咖啡酸-O-甲基转移酶(caffeic acid-O-methyltransferase, COMT)负责木质素等物质的生物合成,对植物的生长发育具有重要作用,在辣椒中COMT还参与辣椒素的合成。利用生物信息学方法在辣椒全基因组中共鉴定出58个COMT家族成员,其不均匀地分布在11条染色体上,编码的氨基酸数在99~372 aa之间,分子量介于10.89~42.04 kDa,大部分被定位在细胞质基质中。系统进化分析将COMT家族成员分为3个组,同一组内大多数成员具有相同的外显子和内含子数量,处于同一分支的蛋白序列具有相同或相似的保守基序。物种内共线性结果显示辣椒COMT家族成员只存在一个片段重复事件;辣椒COMT基因与番茄、大豆的共线性对分别为10、5,与水稻的共线性对仅为1,与玉米无共线性对。顺式作用元件分析表明,辣椒COMT基因中存在大量激素响应元件、胁迫响应元件、生长发育相关元件。辣椒COMT基因与miRNA的相互作用分析发现32条Ca COMT序列与来自mi R396、miR397等多个家族的29个miRNA有潜在的靶向关系。不同组织和果实发育阶段的表达模式发现,部分COMT基因在根、茎、叶、花蕾、花、果实中均不表达或表达量较低,有的COMT基因只在特定组织中表达,表明COMT的表达具有时空特异性。辣椒COMT家族基因在低温胁迫和盐胁迫响应过程中的表达模式分析发现, CaCOMT13、CaCOMT37、CaCOMT46可能同时参与低温胁迫和盐胁迫。Caffeic acid O-methyltransferase(COMT)is essential for the biosynthesis of lignin and plays a significant role in plant growth and development.In Capsicum annuum,COMT also involves in the synthesis of capsaicin.A total of 58 members of the COMT gene family were identified in pepper genome using bioinformatics methods.These genes were found to be unevenly distributed across 11 chromosomes,with their amino acid lengths varying from 99 to 372 and molecular weights ranging from 10.89 to 42.04 kDa.Predominantly,these proteins were localized in the cytoplasmic matrix.Phylogenetic analysis categorized the COMT family members into three distinct groups,with most members within the same group sharing a similar number of exons and introns,as well as conserved motifs in protein sequences that clustered on the same phylogenetic branch.Intraspecific collinearity analysis revealed a single fragment duplication event within the pepper COMT gene family.Comparative collinearity analysis with other species showed that pepper had 10 collinear COMT gene pairs with tomato,5 with soybean,and only 1 with rice,while no collinear pairs were found with maize.The investigation of cis-acting regulatory elements indicated an abundance of plant growth,hormone-responsive,and stress-responsive elements within the capsicum COMT genes.The interaction between pepper COMT gene and miRNA suggests the discovery of potential targeting relationships between 32 CaCOMT sequences and 29 miRNAs from multiple families including miR396 and miR397.Expression pattern analysis across different tissues and fruit developmental stages revealed that some COMT genes were not expressed or expressed at low levels in roots,stems,leaves,flower buds,flowers,and fruits,while certain COMT genes exhibited tissue-specific expression,suggesting spatiotemporal regulation of COMT gene expression.The analysis of expression patterns of pepper COMT family genes under cold and salt stress revealed that CaCOMT13,CaCOMT37,and CaCOMT46 may involve in both cold stress and salt stress r
