摘 要:介绍了一种利用多功能植物效率分析仪M-PEA同步测量两个光系统在光下的实际光化学效率的方法。通过测量光适应下的稳态叶绿素荧光和最大叶绿素荧光,然后计算得出PSⅡ的实际光化学效率(Φ_(PSⅡ))。结合暗适应的最大叶绿素荧光还可以计算非光化学猝灭(NPQ)。完全光适应后,Φ_(PSⅡ)随光强增强显著下降,NPQ则显著升高。光适应下820 nm光反射动力学曲线的初始下降速率反映了PSⅠ的实际光化学效率(Φ_(PSⅠ))。随光强增加,Φ_(PSⅠ)显著下降。研究还表明,与高温强光对Φ_(PSⅡ)影响较大不同,低温弱光对Φ_(PSⅠ)的影响更大。用M-PEA测定的Φ_(PSⅡ)与调制式叶绿素荧光仪测定的Φ_(PSⅡ)呈直线正相关。总之,本方法可以用于研究环境胁迫或生长发育等对光合机构的影响。Using a Multifunctional Plant Efficiency Analyzer M-PEA,a method to simultaneously measure the actual photochemical efficiency of two photosystems under light conditions was introduced.Though measuring steady-state chlorophyll fluorescence and the maximum chlorophyll fluorescence under light adaptation conditions,the actual photochemical efficiency of PSII(Φ_(PSⅡ))could calculated.Combined with the maximum chlorophyll fluorescence in dark adaptation,non-photochemical quenching(NPQ)could also be calculated.After complete light adaptation,Φ_(PSⅡ)significantly decreased with the increase of light intensity,while NPQ significantly increased.The initial decrease rate of the 820 nm light reflection kinetics under light adaptation conditions reflected the actual photochemical efficiency of PSI(Φ_(PSⅠ)).With the increase of light intensity,Φ_(PSⅠ)significantly decreased.It was also showed that,differed from high temperature coupled with high light which showed more effect onΦ_(PSⅡ),low temperature coupled with low light exhibited more effect onΦ_(PSⅠ).TheΦ_(PSⅡ)measured with M-PEA showed linear positive correlation with that measured with a modulated chlorophyll fluorescence analyzer.In conclusion,the method can be used to study the effect of environmental stress or growth and development on the photosynthetic apparatus.
