摘 要:【目的】低温是影响水稻生长、发育和生产的环境因素,鉴定水稻耐冷新基因,揭示耐冷新机制是提高水稻耐冷性的重要途径。【方法】以苗期低温处理0、12、24、36 h的耐冷水稻品种云科粳1号为试验材料,利用转录组测序技术筛选与低温胁迫紧密相关的代谢通路和差异表达基因。【结果】共获得云科粳1号转录组数据76.16 Gb,与参考基因组位置的比对效率达93.56%。基因表达数量韦恩图显示,低温胁迫12、24、36 h后有781个基因共表达。三个比较组合(L_(2)vs L_(1)、L_(3)vs L_(1)、L_(4)vs L_(1))差异基因统计分析显示,L_(2)vs L_(1)差异基因数为6444个,L_(3)vs L_(1)差异基因数为10269个,L_(4)vs L_(1)差异基因数为12576个。不同比较组合基因GO功能富集将差异基因分为生物过程、细胞组分和分子功能三大类,其中,L_(2)vs L_(1)差异基因生物过程、细胞组分和分子功能占比为63.11%、12.25%、24.64%,L_(3)vs L_(1)差异基因各自占比为62.57%、13.54%,23.89%,L_(4)vs L_(1)差异基因各自占比为62.71%、13.69%、23.60%;KEGG代谢通路富集分析表明,植物激素信号转导代谢、植物MAPK信号通路、甘油磷脂代谢通路等参与响应低温胁迫过程。实时荧光定量分析表明,MYB转录因子、WRKY转录因子、蛋白磷酸酶7个参与低温胁迫响应相关的差异表达基因qRT-PCR表达模式与RNA-seq的一致。【结论】云科粳1号具有较强的耐冷性,GO和KEGG代谢通路分析表明,植物MAPK信号、植物激素信号转导及甘油磷脂代谢等均参与响应低温胁迫过程。本研究为水稻耐冷基因克隆及耐冷机制解析提供了理论依据。【Objective】Low temperature is an important environmental factor that affects the growth,development,and production of rice.Identifying new genes for cold tolerance in rice and revealing new mechanisms of cold tolerance are important ways to improverice cold tolerance.【Method】The cold-tolerant variety Yunkejing 1 was chosen as the experimental material.After low temperaturetreatment for 0,12,24,and 36 hours,transcriptome sequencing technology was utilized to identify metabolic pathways anddifferentially expressed genes associated with cold stress.【Result】A total of 76.16 Gb of transcriptome data were obtained forYunkejing 1.The Venn plot of gene expression showed that 781 genes were co-expressed after 12,24,and 36 hours of low temperature.Statistical analysis of three comparative combinations of differentially expressed genes showed that the L_(2)vsL_(1) comparativecombination had 6444 differentially expressed genes,the L_(3)vL_(1) comparative combination had 10269 differentially expressed genes,and the L_(4)vsL_(1) comparative combination had 12576 differentially expressed genes.Gene GO functional enrichment of differentcomparative combinations divides differentially expressed genes into three categories:biological processes,cellular components,andmolecular functions,the DEGs of L_(2)vsL_(1)was accounted for 63.11%,12.25%and 24.64%,the DEGs of L_(3)vsL_(1) was accounted for62.57%,13.54%and 23.89%%,the DEGs of L_(4)vsL_(1)was accounted for 62.71%,13.69%and 23.6%respectively.KEGG metabolicpathway enrichment analysis showed that plant hormone signal transduction metabolism,plant MAPK signaling pathway,andGlycerophospholipid metabolism were involved in the response to low temperature stress process.The expression patterns of 7 DEGs,including MYB transcription factor,WRKY transcription factor,Protein phosphatase and ethylene response factor gene,might berelated to low temperature stress response by Real-time PCR analysis were consistent with those of RNA-Seq.【Conclusion】Yunkejing 1 has strong cold tole
