摘  要:【目的】鉴定并分析苹果NHX基因家族的生物学信息、盐碱胁迫下的基因表达及其功能。【方法】利用氨基酸特异序列和关键词在苹果基因组中检索到17个NHX基因家族候选成员;通过Pfam数据库和SMART数据库最终确认10个苹果NHX基因家族成员,并分别命名为MdNHX1~MdNHX10;利用TBtools、MEGA-X、PlantCARE和MEME等软件对这10个苹果NHX基因家族成员做进一步分析,并利用RNA-seq测定MdNHX基因在盐碱胁迫下的表达情况。【结果】MdNHX基因编码蛋白质的氨基酸数为539~634 aa;分子量为59278.40~69769.84 Da;等电点为5.08~8.99,其中,只有MdNHX2和MdNHX4的等电点小于7,为酸性蛋白,其他成员的等电点均大于7,为碱性蛋白;不稳定系数为33.44~45.41,其中,MdNHX1、MdNHX4、MdNHX7和MdNHX8的不稳定系数小于40,属于稳定蛋白,其他成员均为不稳定蛋白;脂肪系数为101.48~115.52;平均亲水系数为0.364~0.583,均大于0,因此MdNHX可能均为疏水蛋白;它们具有10~11个跨膜螺旋结构,且均定位于液泡膜上;染色体定位和复制事件分析显示有4对MdNHX基因存在片段重复,这4对MdNHX同源基因的Ka/Ks值均小于1,表明苹果NHX基因家族进化过程存在较强的纯化选择;在盐碱胁迫下,多数MdNHX基因的表达被上调;蛋白相互作用分析显示,MdNHX蛋白主要与1-吡咯啉-5-羧酸合成酶、水孔蛋白、质膜ATP酶、乙醛脱氢酶等存在相互作用;GO功能分析表明MdNHX可能在调节苹果K^(+)和Na^(+)转运及盐碱胁迫响应中发挥重要作用。【结论】在苹果基因组GDDH13 v1.1中检索并鉴定得到10个完整的苹果NHX基因家族成员,进一步研究分析发现,苹果NHX基因家族成员可能在植物对胁迫、光以及植物激素的响应中发挥重要作用。[Objective]The present article aimed to identify and analyze the biological information of the apple NHX gene family,gene expression and function under salt alkali stress.[Method]Using amino acid specific sequences and keywords,17 candidate members of the NHX gene family were retrieved from the apple genome;Then,10 members of the apple NHX gene family were finally confirmed through the Pfam database and SMART database,and were named MdNHXI to MdNHXIO respectively;Finally,further analysis was conducted on these 10 members of the apple NHX gene family using software such as TBtools,MEGA-X,PlantCARE and MEME,and RNA-seq was used to determine the expression of the MdNHX gene under salt alkali stress.[Result]The number of amino acids encoded by these MdNHX genes was 539-634 aa;The molecular weight ranged from 59278.40 to 69769.84 Da;The isoelectric point was 5.08-8.99,among which only MdNHX2 and MdNHX4 had an isoelectric point less than 7,indicating acidic proteins,while the isoelectric points of other members were all greater than 7,indicating alkaline proteins;The instability coefficient ranged from 33.44 to 45.41,among which MdNHX1,MdNHX4,MdNHX7 and MdNHX8 had instability coefficients less than 40 and belonged to stable proteins,while the other members were all unstable proteins;The fat coefficient ranged from 101.48 to 115.52;The average hydrophilicity coefficient ranged from 0.364 to 0.583,all of which were greater than O,indicating that MdNHX may be hydrophobic proteins;They had 10-11 transmembrane spiral structures and were all located on the vacuolar membrane;Chromosome localization and replication event analysis showed that there were 4 pairs of MdNHX genes with fragment duplication,and the Ka/Ks values of these 4 pairs of MdNHX homologous genes were all less than 1,indicating strong purificationselection in the evolutionary process of the apple NHX gene family;Under salt alkali stress,the expression of most MdNHX genes was upregulated;Protein interaction analysis showed that MdNHX protein mainly interacted w
