摘 要:【目的】更好地了解吴茱萸转录本基因信息,为吴茱萸有效成分生物合成途径的阐明奠定基础。【方法】提取吴茱萸果实4个时期总RNA,开展混合样三代全长转录组测序,对获得的转录本进行功能注释和分类。【结果】获得了73 Gb的原始数据,经聚类及校正后得到63744条一致性序列,高质量序列达63550条,达99.7%,去冗余后最终获得19044条Unigene。与各数据库进行比对后,有18704条Unigene被注释,总注释率为98.21%,其中在NR数据库中注释最多,达18662条,注释最少的为KEGG数据库,仅有5943条。【结论】对吴茱萸有效成分的生物合成相关基因进行分析,发现有月桂烯、罗勒烯及芳樟醇等挥发性萜类成分53条候选基因;柠檬苦素生物合成途径发现2条候选基因(不含MVA及MEP途径);吴茱萸碱、吴茱萸次碱生物合成途径发现53条候选基因,共计108条基因。[Objective]The present paper aimed to better understand the gene information of Tetradium ruticarpum transcripts and lay a foun-dation for elucidating the biosynthetic pathway of effective components of T.ruticarpum.[Method]We extracted total RNA from the fruits of T.ruticarpum at four different developmental stages,carried out mixed sample third-generation full-length transcriptome sequencing,and per-formed functional annotation and classification on the obtained transcripts.[Result]73 Cb raw data were obtained.After clustering and cor-rection,63744 consistent sequences were obtained,and the high-quality sequences reached 63550,reaching 99.7%.After redundancy re-moval,19044 Unigenes were finally obtained.After compared with various databases,18704 Unigenes were annotated,with a total annotation rate of 98.21%.Among them,the NR database had the most annotations,reaching 18662,and the KEGG database had the least annotations,only 5943.[Conclusion]An analysis of genes related to the biosynthesis of active components in T.ruticarpum was conducted,revealing 53 candidate genes for volatile terpenoid components such as myrcene,ocimene,and linalool;2 candidate genes were identified in the biosyn-thetic pathway of limonin(excluding MVA and MEP pathways);and 53 candidate genes were found in the biosynthetic pathways of rutae-carpine and evodiamine,totaling 108 genes.
