摘 要:【目的】对蛇足石杉SAUR50基因家族的分子生物信息学及组织特异性表达进行分析,为提高蛇足石杉产量提供参考依据。【方法】基于蛇足石杉全长转录组数据,利用生物信息学的方法对蛇足石杉SAUR50基因家族成员进行鉴定,分析其蛋白的理化性质、亚细胞定位、结构域和保守基序,对蛇足石杉SAUR50基因家族成员组织表达特性进行分析。【结果】共鉴定出11个SAUR50基因家族成员(HsSAUR50-1~HsSAUR50-11),编码的氨基酸数量为62~138个,理论等电点为6.71~10.31,其蛋白分子量为7290.48~16258.16 kD,多为碱性蛋白,不具有跨膜结构,且均为稳定亲水性蛋白。蛇足石杉SAUR50蛋白二级结构均由α螺旋、β折叠、延伸链和无规则卷曲4种结构共同组成,有蛋白结合、转移酶链和DNA蛋白结合3种三级结构,亚细胞定位在叶绿体、细胞核中。系统发育进化分析将蛇足石杉SAUR50蛋白家族成员分为两大类(Ⅰ和Ⅱ类),其中Ⅰ类可分为2个亚类。蛋白结构域分析结果显示,SAUR50家族蛋白共有2种结构域,分别为Auxin inducible和Auxin inducible superfamily,具有促进基因在生长素水平表达的功能。分析蛇足石杉SAUR50家族蛋白序列共获得3种保守基序,其中motif 1在遗传进化中趋于稳定。蛇足石杉SAUR50基因家族成员有与生长发育、激素响应、抗逆等相关的17种顺式作用元件。HsSAUR50-8等6个基因在茎中高表达,HsSAUR50-2和HsSAUR50-6基因表达量排序均为茎>叶>芽胞>孢子。【结论】共鉴定出11个蛇足石杉SAUR50基因家族成员,具有高度保守基序和功能结构域,其组织表达具有特异性。蛇足石杉SAUR50基因家族大部分成员可能主要参与蛇足石杉茎和叶的发育调控,能促进蛇足石杉植株增高及茎叶生长发育。[Objective]The molecular bioinformatics and tissue-specific expression of the SAUR50 gene family of Huperzia serrata were ana-lyzed to provide reference for improving the yield of H.serrata.[Method]Based on the full-length transcriptome data of H.serrata,bioinfor-matics was used to identify the members of the SAUR50 gene family,analyze the physicochemical properties,subcellular localization,domain and conserved motifs of the proteins,and analyze the tissue expression properties of the SAUR50 gene family members.[Result]A total of 11 HsSAUR50 gene family members were identified(HsSAUR50-1-HsSAUR50-11).The encoded protein contained 62-138 amino acids,the i-soelectric point was 6.71-10.21,and the relative molecular weight was 7290.48-16258.16 kD.Most of them were alkaline proteins,and did not have a transmembrane structure and were stable and hydrophilic proteins.The secondary structure of H.serrata SAUR50 protein was composed of four structures:alpha helix,beta turn,extended strand,random coil.The SAUR50 protein had three tertiary structures:protein binding,transferase chain and DNA protein binding.Subcellular was localized in chloroplast and nucleus.Phylogenetic analysis showed that the members of the SAUR50 protein family members were divided into two groups(I and II),and the I group could be divided into two sub-groups.The results of protein domain analysis showed that the SAUR50 family proteins had two domains,Auxin inducible and Auxin induc-ible superfamily,which had the function of promoting gene expression at the auxin level.The SAUR50 family protein sequence analysis of H.serrata obtained three conserved motifs,of which motif 1 tended to be stable in genetic evolution.SAUR50 gene family members had 17 cis-acting elements related to growth and development,hormone response,stress resistance and so on.The expression level of six genes such as HsSAUR50-8 was the highest in stems,and the expression levels of HsSAUR50-2 and HsSAUR50-6 genes were stems>leaves>gemma>spores.[Conclusion]A total of1 SAUR50 gene family
