摘 要:Background:Trachelospermi Caulis et Folium, the stems and leaves of Trachelospermum jasminoides(Lindl.)Lem,has been used as a remedy for rheumatism, gonarthritis, backache and pharyngitis, according to the Chinese Pharmacopoeia. However, the corresponding chemical substances of this ethnic medicinal plant with anti-inflammatory activity kept unclear. This study aimed to explore the active substances attribute to its anti-rheumatic activity of this medical plant.Methods:Ultra-high performance liquid chromatography combined with quadrupole time-of-flight mass spectrometry was used to identify the chemical components in the extract of T.jasminoides.Molecular docking was applied to analyze the binding affinity of potential active lignans to inducible nitric oxide synthase enzyme. The anti-inflammatory effects of the potential active fraction were verified by cell experiments in vitro.Results:Forty-one compounds, mainly are lignans and nine of which were reported for the first time from the genus of Trachelospermum, were characterized using ultra-high performance liquid chromatography quadrupole-time-of-flight mass spectrometry method.Molecular docking results showed these identified lignans had excellent interactions with the inducible nitric oxide synthase enzyme, and the lignans-enriched fraction could significantly inhibit the release of nitric oxide and the expression of inducible nitric oxide synthase protein in LPS-induced RAW264.7 cells. Conclusion:Lignans are the main active substances that responsible for the anti-inflammatory activity of the ethnic herbal medicine T.jasminoides.
