摘 要:目的探讨急性/慢性低压缺氧对大鼠小肠中段组织的影响。方法本研究将模拟高原低压缺氧环境对大鼠小肠中段组织进行了转录组学检测。我们将大鼠随机分为3组:正常对照组(S),急性低压缺氧组(W2S)即暴露于低压缺氧环境2周和慢性低压缺氧组(W4S)即暴露于低压缺氧环境4周。随后对3组大鼠的小肠中段组织进行了RNA谱测序分析。结果主成分分析显示,W4S组和W2S组分别与对照组(S)相比均存在显著差异,共鉴定出266个差异表达基因。京都基因与基因组百科全书(Kyoto encyclopedia of gene and genome,KEGG)KEGG通路主要富集到蛋白质消化与代谢途径、神经活性配体-受体相互作用通路、视黄醇代谢、葡萄糖醛酸代谢、抗坏血酸和激素等生物合成代谢等5种途径。通过共表达分析发现,神经活性配体与受体的相互作用途径和补体及凝血途径在暴露于低压缺氧环境4周(W4S组)显著上调,结果与KEGG分析一致。结论此研究主要利用RNA测序(RNA-seq)技术探讨大鼠小肠中段组织在高原低压低氧恶劣环境中基因表达的变化,为肠道应对高原环境中的适应性改变提供新的证据。Objective To investigate the effects of acute/chronic low-pressure hypoxia on rat middle small intestinal tissues.Methods In this study,we performed transcriptomic assays on rat middle small intestine tissues in a simulated plateau low-pressure hypoxic environment.We randomly divided the study rats into three groups:normal control group(S),acute low-pressure hypoxia group(W2S)which was exposed to low-pressure hypoxic environment for 2 weeks,and chronic low-pressure hypoxia group(W4S)which was exposed to low-pressure hypoxic environment for 4 weeks.RNA profiling was subsequently performed on the middle small intestinal tissues of all three groups of rats.Results total of 266 differentially expressed genes were identified in study tissues.Furthermore,the principal component analysis showed that the W4S and W2S groups were significantly different from the control group.KEGG pathway was mainly enriched with five signaling pathways,including protein digestion and metabolism,neuroactive ligand-receptor interaction pathway,retinol metabolism,glucuronide metabolism,ascorbic acid and hormone biosynthesis metabolism.The co-expression analysis revealed that the neuroactive ligand-receptor interaction pathway and the complement and coagulation pathways were significantly up-regulated by exposure to low pressure hypoxia for 4 weeks(W4S group),and the results were consistent with KEGG analysis results.Conclusion This study mainly used RNA sequencing(RNA-seq)technology to explore changes of gene expressions in middle rat small intestine tissues under the harsh environment of low pressure and hypoxia in the plateau.Our results provide novel evidences for the adaptive changes in the intestinal tract in response to the plateau environment.
