摘  要:在形态学初步鉴定的基础上,使用DNA条形码技术对山东省多地蔬菜上的蓟马进行检测,以采集的样本为靶标,利用DNA条形码通用型引物扩增其线粒体DNA细胞色素C氧化酶亚基Ⅰ(mtDNA COⅠ)基因(约650 bp),通过测序与比对分析,以邻接法构建系统发育树,并以Kimura双参数模型计算种内、种间遗传距离。采自山东省的20种蔬菜蓟马,种间平均遗传距离为0.197,种内平均遗传距离为0.024,其中以苏丹呆蓟马(Anaphothrips sudanensis)的种内遗传距离最大,为0.087。经序列比对和系统发育树分析,初始的形态学鉴定结果可靠,丰富了山东省蔬菜蓟马种类调查的基础数据。This study utilizied morphological identification with DNA barcoding technology to detect and analyze thrips in vegetable crops in Shandong,with the aim of rapidly and accurately identifying thrips species.The universal DNA barcode primer was used to to amplify the mitochondrial cytochrome c oxidase subunit I gene(approximately 650 bp)the thrips samples collected in Shandong Province.Through sequencing and comparison analysis of the base sequence of the target fragment,the phylogenetic tree was constructed using the adjacency method(NJ method),and the Kimura's two parameter model was used to calculate the intraspecific and interspecific genetic distances.The average genetic distance between 20 species of vegetable thrips in Shandong Province was found to be 0.197,and the average intraspecific genetic distance was 0.024.The intraspecific genetic distance of Anaphothrips sudanensis was found to be the maximum,at 0.087.The results of the sequence alignment and phylogenetic tree analysis showed that the initial morphological identification results of vegetable thrips were reliable and could be used as the basic data for the investigation of vegetable thrips in Shandong Province.
