摘 要:目的研究舒脑欣滴丸对细胞缺氧损伤的保护作用,并初步探讨其可能影响的作用环节,为高原缺氧损伤防护药物的研发提供依据。方法建立PC12细胞和H9c2细胞缺氧模型,给予复方丹参滴丸与不同剂量舒脑欣滴丸干预,设置正常对照组、缺氧模型组、复方丹参滴丸阳性对照组(125.00μg/ml复方丹参滴丸溶液)、舒脑欣低剂量组(7.81μg/ml舒脑欣滴丸溶液)、舒脑欣中剂量组(31.25μg/ml舒脑欣滴丸溶液)、舒脑欣高剂量组(125.00μg/ml舒脑欣滴丸溶液),检测各组的细胞乳酸脱氢酶(LDH)释放量、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性,采用Hoechst/PI染色检测细胞凋亡,根据细胞损伤程度评价舒脑欣滴丸对细胞缺氧的保护作用,采用Western Blot检测PC12细胞和H9c2细胞组蛋白去乙酰化酶2(HDAC2),以及核因子κB(NF-κB)、NF-κB抑制因子(IκB)磷酸化活性表达水平。结果①舒脑欣中剂量组、舒脑欣高剂量组的PC1细胞与2H9c2细胞的LDH释放量、MDA含量均低于缺氧模型组,SOD活性均高于缺氧模型组,差异有统计学意义(P<0.05)。②舒脑欣中剂量组、舒脑欣高剂量组的PC12细胞与H9c2细胞凋亡率均低于缺氧模型组与复方丹参滴丸阳性对照组,差异有统计学意义(P<0.05)。③舒脑欣低剂量组、舒脑欣中剂量组、舒脑欣高剂量组PC12细胞与H9c2细胞的HDAC2表达量高于缺氧模型组,磷酸化NF-κB(p-NF-κB)p65、磷酸化IκB(p-IκB)表达量低于缺氧模型组,差异有统计学意义(P<0.05)。舒脑欣低剂量组、舒脑欣中剂量组、舒脑欣高剂量组PC12细胞的p-NF-κB p65表达量与H9c2细胞的p-IκB表达量均低于复方丹参滴丸阳性对照组;舒脑欣高剂量组PC12细胞与H9c2细胞的HDAC2表达量高于复方丹参滴丸阳性对照组,p-IκB表达量低于复方丹参滴丸阳性对照组;舒脑欣低剂量组、舒脑欣中剂量组H9c2细胞的p-NF-κB p65表达量均高于复方丹参滴丸阳性对照组,差异�Objective To study the protective effect of Shunaoxin Dropping Pills on cell hypoxic injury,and to explore the possible links of action,so as to provide a basis for the research and development of protective drugs against hypoxic injury at high altitude.Methods Hypoxic models of PC12 cells and H9c2 cells were established and treated with Compound Danshen Dropping Pills and different doses of Shunaoxin Dropping Pills.The normal control group,hypoxia model group,positive control group of Compound Danshen Dropping Pills(125.00μg/ml Compound Danshen Dropping Pills solution),low dose of Shunaoxin group(7.81μg/ml Shunnaoxin Dropping Pills solution),medium dose of Shunaoxin group(31.25μg/ml Shunnaoxin Dropping Pills solution),and high dose of Shunaoxin group(125.00μg/ml Shunnaoxin Dropping Pills solution)were set up.The release of lactate dehydrogenase(LDH),the content of malondialdehyde(MDA)and the activity of superoxide dismutase(SOD)were detected among the groups.Hoechst/PI staining was used to detect cell apoptosis.Western Blot was used to detect the expression of histone deacetylase 2(HDAC2),phosphorylation activity of nuclear factor-κB(NF-κB)and inhibitor of NF-κB(IκB)in PC12 cells and H9c2 cells.Results①The LDH release and MDA content of PC12 cells and H9c2 cells in the medium dose of Shunaoxin group and the high dose of Shunaoxin group were lower than those in the hypoxia model group,and the SOD activity were higher than that in the hypoxia model group,with statistically significant differences(P<0.05).②The apoptosis rates of PC12 cells and H9c2 cells in the medium dose of Shunaoxin group and high dose of Shunaoxin group were lower than those in the hypoxia model group and the positive control group of Compound Danshen Dropping Pills,and the differences were statistically significant(P<0.05).③The expression of HDAC2 in PC12 cells and H9c2 cells of low dose of Shunaoxin group,medium dose of Shunaoxin group and high dose of Shunaoxin group were higher than those of hypoxia model group,the expression
