摘 要:高纯度的单一对映异构体在生命的产生和进化过程中至关重要,因此,对映异构体的手性分离在生命科学和制药工业中都具有重要意义。色谱分离是广泛使用的手性分离策略。得益于高效和多样化手性固定相的开发,色谱过程特别是模拟移动床色谱过程成为实现制备级手性拆分的一种有效方法。溶剂组成对吸附强度、溶解度、扩散系数都有显著影响,也会导致固定相结构的变化。探讨了流动相组成对色谱拆分关键工艺参数的影响。模型体系为阿折地平对映体,温度为298 K,固定相为ChiralPak AD,流动相为正己烷和乙醇的混合物,乙醇体积比为20V%~40V%。High-purity single enantiomers are essential in the generation and evolution of life,so chiral separation of enan⁃tiomers is of great significance in both the life sciences and pharmaceutical industries.Chromatographic separation is a widely used chiral separation strategy.Thanks to the development of highly efficient and diverse chiral stationary phases,chromatographic processes,especially those that simulated moving bed chromatography,have become an effective method for achieving preparative-grade chiral resolutions.Solvent composition has a significant effect on adsorption strength,solu⁃bility,diffusion coefficient,and also leads to changes in the structure of the stationary phase.Here,we discuss the influ⁃ence of mobile phase composition on key process parameters for chromatographic resolution.The model system was an enan⁃tiomer of Azelnidipine at a temperature of 298 K,the stationary phase was ChiralPak AD,and the mobile phase was a mix⁃ture of n-hexane and ethanol with an ethanol volume ratio of 20V%~40V%.
