不同pH及Cd^(2+)浓度下文蛤肝胰腺的氧化应激效应

摘  要:用不同pH的Cd^(2+)溶液对文蛤(Meretrix meretrix)进行染毒,测定不同染毒时间(2天、4天、6天)文蛤肝胰腺对Cd^(2+)的富集量,分析Cd^(2+)对肝胰腺细胞超微结构的影响;并测定了肝胰腺中H_(2)O_(2)、MDA和NO含量及抗氧化酶(CAT、SOD、GPx)活性和金属硫蛋白(MT)mRNA表达量.结果显示,Cd^(2+)在文蛤肝胰腺中的富集量及H_(2)O_(2)、MDA、NO含量均随Cd^(2+)浓度、染毒时间的增加而上升,pH 7.0组的肝胰腺中Cd^(2+)的富集量及MDA含量均高于pH 8.3组.随着染毒时间增长和染毒浓度提高,细胞膜性结构受损越来越严重.抗氧化酶CAT、SOD、GPx活性在6 mg/L Cd^(2+)浓度组中,染毒时间较短时其活性较高,且pH 7.0组的抗氧化酶活性高于pH 8.3组;但随着Cd^(2+)浓度和染毒时间的增加,pH 7.0组活性下降较pH 8.3组更显著.相同染毒条件下的MT mRNA表达量随染毒时间的延长总体呈下降趋势,相同染毒时间下6 mg/L Cd^(2+)浓度组的MT mRNA表达量显著高于其他组,且pH 8.3组高于pH 7.0组.分析了pH值和Cd^(2+)对文蛤肝胰腺的氧化损伤生物标志物的效应,有助于丰富海洋酸化与重金属毒理作用在海洋贝类中的研究内容.Meretrix meretrix was exposed to different pH and Cd^(2+)concentration.The level of Cd^(2+)in the hepatopancreas of M.meretrix was measured after different exposure time(2,4,6 days),and the effects of Cd^(2+)on the ultrastructure of hepatopancreas cells were analyzed.Besides,the levels of H_(2)O_(2),MDA and NO,the activities of antioxidant enzymes(CAT,SOD,GPx)and the expression of MT mRNA of hepatopancreas in the clam were also determined.The results showed that the levels of Cd,H_(2)O_(2),MDA and NO in the hepatopancreas of M.meretrix increased with the increasing Cd^(2+)concentration and exposure time.The levels of Cd 2 and MDA in hepatopancreas under pH 7.0 were higher than those under pH 8.3.With the increase of exposure time and Cd^(2+)concentration,the damage of cell membrane structure became more and more serious.The activities of CAT,SOD and GPx were higher at 6 mg/L Cd^(2+)concentration and short exposure time,and the activities of antioxidant enzymes under pH 7.0 were higher than that under pH 8.3.However,with the increase of Cd^(2+)concentration and exposure time,the activities of CAT,SOD and GPx at pH 7.0 decreased more than that of pH 8.3.The expression level of MT mRNA under the same exposure conditions showed a decreasing trend with the prolongation of the exposure time.At the same time,the level of MT mRNA expression in the 6 mg/L Cd^(2+)group was significantly higher than that in other groups,and the expression of MT mRNA under pH 8.3 was higher than that under pH 7.0.The comprehensive effects of pH and Cd^(2+)on oxidative damage biomarkers in the hepatopancreas of M.meretrix were analyzed in this paper,which is helpful to enrich the research content of ocean acidification and toxicity of heavy metals in marine shellfish.
