摘  要:为研究离子细胞(Ionocytes)的顶端开口(Apical opening)大小受何种因素影响,本研究以斑马鱼为模式生物,利用免疫荧光和药物处理的方法探究影响顶端开口的因素。本研究中,发现了一种紧密连接蛋白Claudin-5的抗体,它能有效标记斑马鱼胚胎离子细胞的轮廓及其顶端开口,解决了目前对离子细胞开口的表征研究多依赖于操作较为繁琐的电镜的问题。进一步地,通过小分子药物处理,发现离子细胞顶端开口的大小受到钙离子及非肌肉肌球蛋白的控制。具体而言,钙离子抑制剂处理胚胎会导致离子细胞顶端开口缩小,非肌肉肌球蛋白Ⅱ的抑制也会出现类似的现象。而用小分子(如肾上腺素和甲状腺素)处理则对离子细胞顶端开口大小并无明显影响。本研究不仅提供了一个可以用来标记并监测离子细胞顶端开口的方案,还发现了钙离子和非肌肉肌球蛋白Ⅱ对离子细胞顶端开口大小的调控功能,这些研究有助于更深入地理解离子细胞结构的稳态维持和生理功能的行使。In order to study the factors affecting the size of the apical opening of ionocytes,this study used zebrafish as a model organism to explore the factors by immunofluorescence and drug treatment.The apical opening was usually investigated via electron microscopy,which is relatively laborious.In this study,we reported a simple method to visualize apical opening of ionocytes via antibody staining of Claudin-5,a tight junction protein.We analyzed the apical opening of ionocytes using several small molecule compounds.Our data showed that treatment of zebrafish embryos with calcium inhibitors reduced the size of apical opening in ionocytes.Similarly,incubation with inhibitor of non-muscle myosinⅡalso resulted in the decreased size of apical opening.In contrast,treatment with either epinephrine or thyroid hormones had no effect on the apical opening of ionocytes.These results suggested that the apical opening of ionocytes is controlled by a calcium dependent regulatory mechanisms which may require the involvement of non-muscle myosinⅡ.Our data may provide further information for the study of the homeostasis and physiological function of ionocytes.
