摘 要:为探究在不同盐度条件下长期养殖对驼背鲈(Cromileptes altivelis)幼鱼基因表达的影响,本研究将体质量1.1~1.7 g的驼背鲈幼鱼分别养殖于盐度为32的自然海水和盐度为18的低盐度海水中,分别在养殖第50和60天时选取不同盐度组驼背鲈幼鱼的脑、肝脏和肌肉组织进行转录组分析,以筛选差异表达基因(Differentially expressed genes,DEGs)。研究结果表明:脑组织中的差异基因显著多于肝脏和肌肉组织;脑组织中差异基因主要参与细胞分裂、营养物质代谢以及细胞周期等生物学过程;肝脏组织中差异基因主要参与糖类物质代谢、氨基酸代谢以及酶复合体形成等过程;肌肉组织中的差异基因则主要与细胞生长发育和分裂有关,并与生长相关的信号通路如丝裂原活化蛋白激酶(Mitogen-activated protein kinase,MAPK)信号通路、钙离子信号通路、Wnt信号通路以及胰岛素信号通路等密切相关。本研究还采用实时荧光定量PCR(Quantitative real-time PCR,qRT-PCR)对3个组织中分别随机选取的3个DEG进行表达量验证。结果显示,这些基因的表达趋势与转录组测序的结果一致,从而证明测序数据可信。本研究的结果为今后继续开展驼背鲈养殖育种工作和盐度适应的分子机制提供了参考。This study investigated the effects of long-term aquaculture under different salinity conditions on gene expression of the juvenile Cromileptes altivelis,the brain,liver,and muscle tissues rared in low salinity sea water were selected for transcriptome analysis to explore the mechanism of salinity stress on fish.Juveniles with body weight of 1.1~1.7 g were rared in natural sea water with salinity of 32 as control and salinity of 18 as low lalinity group,respectively.Then,the brain,liver,and muscle tissues were collected on days 50 and 60,and the transcriptomes were sequenced by Illumina NovaSeq 6000 platform.Screening conditions for differentially expressed genes(DEGs)were set as p-value<0.05 and|log 2(Fold change)|>1.The results of enrichment analysis showed that the DEGs in brain tissue between the two experimental groups were significantly more than those in liver and muscle tissues.The DEGs in brain tissue were mainly involved in biological processes such as cell division,nutrient metabolism and cell cycle.The DEGs in liver tissue were mainly involved in carbohydrate metabolism,amino acid metabolism,enzyme complex formation and other processes.The DEGs in muscle tissue were mainly related to cell growth and development,division,and growth-related signaling pathways such as MAPK signaling pathway,calcium signaling pathway,Wnt signaling pathway,and insulin signaling pathway.Real-time fluorescence quantitative PCR(qRT-PCR)was used to verify the expression of three DEGs randomly selected from three tissues.The results of this study provide a reference for the future breeding work of Cromileptes altivelis and the molecular mechanism of salinity adaptation.
