摘 要:【目的】研究农抗N2抑制指状青霉与意大利青霉生长的作用机制,以期为由指状青霉与意大利青霉引发的植物病害的生物防治提供材料和理论参考。【方法】通过测定经不同浓度农抗N2处理的指状青霉及意大利青霉的β-1,3-葡聚糖酶活性与几丁质酶活性,研究农抗N2对2种病原真菌细胞壁降解酶活性的影响;通过测定经不同浓度农抗N2处理的指状青霉及意大利青霉的麦角甾醇含量研究农抗N2对病原真菌细胞膜组分的影响;通过测定胞外蛋白质与DNA含量,进一步探究农抗N2对指状青霉与意大利青霉细胞壁与细胞膜完整性的影响。【结果】经农抗N2处理的指状青霉与意大利青霉,其β-1,3-葡聚糖酶与几丁质酶活性在处理24h后显著提高,且酶活性与农抗N2浓度呈正相关。经农抗N2处理的指状青霉与意大利青霉,2种病原真菌细胞膜中麦角甾醇含量在处理24h后显著下降,其麦角甾醇含量与农抗N2浓度呈负相关。在用高浓度N2处理120h后,2种病原真菌的麦角甾醇含量达到最低值,仅为未处理组的16.77%(指状青霉)和16.23%(意大利青霉)。经农抗N2处理的2种病原真菌,胞外蛋白质含量在处理24h后显著增加,并与农抗N2浓度呈正相关;胞外DNA含量在经农抗N2处理后也有不同程度的增加,其含量在高浓度农抗N2处理72h达到最大值,分别为未处理组的1.98倍(指状青霉)与1.35倍(意大利青霉)。【结论】农抗N2能有效诱导病原真菌产生β-1,3-葡聚糖酶与几丁质酶,降解真菌细胞壁中的几丁质与β-1,3-葡聚糖,破坏细胞壁的完整性。此外,农抗N2显著降低病原真菌细胞膜中的麦角甾醇含量,影响麦角甾醇的生物合成,进而破坏细胞膜的完整性。综上,破坏真菌细胞壁与细胞膜的完整性是农抗N2抑制指状青霉与意大利青霉的重要机制。[Objective]This study aims to investigate the action mechanism by which fungicide N2 inhibits the growth of Penicillium digitatum and Penicillium italicum,with the goal of providing materials and theoretical references for the biological control of plant disease caused by these two fungi.[Method]The activities ofβ-1,3-glucanase and chitinase in P.digitatum and P.italicum treated by different concentrations of fungicide N2 were determined in order to investigate the effect of fungicide N2 on the cell wall degradation-related enzymes.The ergosterol content in both fungi treated with different concentrations of fungicide N2 was also determined in order to investigate the effect of fungicide N2 on cell membrane composition in P.digitatum and P.italicum.The effect of fungicide N2 on the integrity of cell wall and membrane was further investigated through determining the extracellular protein and DNA content.[Result]The activities ofβ-1,3-glucanase and chitinase in P.digitatum and P.italicum were significantly increased after after 24 h of treatment with Fungicide,and increased in a concentration-dependent manner.The ergosterol content in both fungi decreased markedly after 24 h treatment with fungicide N2 and exhibited a negative correlation with treatment concentration.The ergosterol content in P.digitatum and P.italicum both reached a minimum value at 120 h,which was 16.77%and 16.23%that of the untreated group,respectively.Moreover,the extracellular protein content in both pathogens increased significantly after 24 h treatment with fungicide N2.It was positively correlated with the treatment concentration.The content of extracellular DNA also increased in different degrees after N2 treatment.The content of extracellular DNA reached the maximum at 72 h after high concentration of N2 treatment,which was 1.98 times(Penicillium digitatum)and 1.35 times(Penicillium italicum)of the untreated group,respectively.[Conclusion]Fungicide N2 could induce the production ofβ-1,3-glucanase and chitinase in P.digitatum and P.ital
