摘  要:目的 分析电针治疗对骨癌痛大鼠镇痛作用及NLRP3通路的影响。方法 选择雄性SPF级SD大鼠30只,分为对照组、模型组和电针组,每组10只。通过检测NLRP3、Caspase-1、IL-1β的蛋白表达水平对3组大鼠疼痛进行评估。结果 在机械性痛阈值上,对照组高于模型组、电针组,电针组高于模型组,差异具有统计学意义(P<0.05);在PWL时间上,对照组高于模型组,且差异具有统计学意义(P<0.05),对照组和电针组对比差异无统计学意义(P>0.05);在自发性疼痛评分上,对照组分值低于模型组、电针组,电针组低于模型组,且差异具有统计学意义(P<0.05);在脊髓组织中的IL-1β、Caspase-1、NLRP3蛋白表达水平上,电针组、模型组和对照组对比差异具有统计学意义(P<0.05);模型组大鼠的脊髓组织中的IL-1β、Caspase-1、NLRP3蛋白表达水平高于对照组和电针组,电针组大鼠的脊髓组织中的IL-1β、Caspase-1、NLRP3蛋白表达水平高于对照组,对比差异具有统计学意义(P<0.05)。结论 电针治疗可有效减轻骨癌痛的痛感,可能与电针刺激疼痛部位NLRP3信号通路激活有关。Objective To analyze the effect of electroacupuncture treatment on the NLRP3 pathway and analgesic effect in rats with bone cancer pain.Methods 30 male SPF SD rats were selected to create a rat tibial cancer pain model,which was divided into three groups,namely the control group,the model group and the electroacupuncture group.All three groups received corresponding intervention for 12 days,and then their pain was reduced.were evaluated,and Western blotting was used to detect the protein expression levels of NLRP3 signaling pathway,namely NLRP3,Caspase-1,and IL-1β.Results On the threshold of mechanical pain,the control group was higher than the model group and the electroacupuncture group,and the electroacupuncture group was higher than the model group,and the difference was statistically significant(P<0.05);In terms of PWL time,the control group was higher than the model group,and the difference was statistically significant(P<0.05).There was no significant difference between the control group and the electroacupuncture group(P>0.05).In terms of spontaneous pain score,the control group scored lower than the model group and the electroacupuncture group,and the electroacupuncture group scored lower than the model group,and the difference was statistically significant(P<0.05).There was a statistically significant difference(P<0.05)in the expression levels of IL-1β,Caspase-1,and NLRP3 proteins in spinal cord tissue between the electroacupuncture group,model group,and control group.The expression levels of IL-1β,Caspase-1,and NLRP3 proteins in the spinal cord tissue of the model group rats were higher than those of the control group and the electroacupuncture group.The expression levels of IL-1β,Caspase-1,and NLRP3 proteins in the spinal cord tissue of the electroacupuncture group rats were higher than those of the control group,and the difference was statistically significant(P<0.05).Conclusions Electroacupuncture treatment can effectively reduce the pain of bone cancer pain,which may be related to the activatio
