摘 要:目的 旨在通过生物信息学手段鉴定含SNW结构域蛋白1(SNW1)在肝细胞癌中的潜在生物标记物价值,并结合细胞实验探索SNW1对肝细胞癌细胞增殖和迁移侵袭能力的影响。方法 通过孟德尔随机化结合生物统计方法鉴定与肝细胞癌具有因果关系的特征基因,采用转录组差异表达和生存分析探索特征基因在肝细胞癌中的表达情况及与预后不良的相关性,确定SNW1可作为潜在生物标记物的潜能并进行基因集富集和免疫细胞浸润的相关性分析。使用实时荧光定量聚合酶链反应检测人肝细胞癌细胞HepG2和Hep3B中SNW1的表达水平。通过转染敲低肝细胞癌细胞中SNW1的表达后,采用细胞计数试剂盒-8、细胞划痕和Transwell实验检测肝细胞癌细胞的增殖、迁移和侵袭能力。结果 SNW1是肝细胞癌的风险基因,在肝细胞癌患者中显著高表达且与患者预后显著相关(P<0.05)。与SNW1显著正相关的通路主要涉及RNA剪接、病毒性癌变与Epstein-Barr病毒感染等生物学过程。SNW1与静止树突细胞和嗜酸性粒细胞呈显著正相关,与调节T细胞和激活自然杀伤细胞呈显著负相关。细胞实验表明敲低SNW1能抑制肝细胞癌细胞的增殖、迁移和侵袭能力。结论 SNW1有潜力成为新的肝细胞癌生物标记物与治疗靶点。Objective To identify the potential biomarker value of SNW domain containing 1(SNW1)in hepatocellular carcinoma through bioinformatics methods,and to explore the effect of SNW1 on the proliferation and invasion abilities of hepatocellular carcinoma cells through cell experiments.Methods Mendelian randomization combined with biostatistical methods were employed to identify characteristic genes with a causal relationship to hepatocellular carcinoma.Transcriptome differential expression and survival analysis were utilized to examine the expression of characteristic genes in hepatocellular carcinoma and the correlation of characteristic genes and prognosis,as well as to identify the potential of SNW1 as a potential biomarker.Additionally,correlation analysis of SNW1 with gene set enrichment and immune cell infiltration were performed.Furthermore,real-time fluorescence quantitative polymerase chain reaction was utilized to ascertain the SNW1 expression in human hepatocellular carcinoma cell lines HepG2 and Hep3B.SNW1 in the hepatocellular carcinoma cells was knocked down by cell transfection,followed by cell counting kit-8,cell scratch,and Transwell assays to detect the proliferation,migration,and invasion abilities of hepatocellular carcinoma cells.Results SNW1 had been identified as a potential risk factor for hepatocellular carcinoma,with significantly elevated expression levels observed in patients(P<0.05).Additionally,SNW1 expression levels were correlated with the prognosis of hepatocellular carcinoma patients(P<0.05).The pathways significantly positively correlated with SNW1 mainly involved biological processes such as RNA splicing,viral carcinogenesis,and Epstein-Barr virus infection.SNW1 was significantly positively correlated with quiescent dendritic cells and eosinophils,and negatively correlated with regulating T cells and activating NK cells.Cell experiments demonstrated that the knockdown of SNW1 inhibited the capacity of hepatocellular carcinoma cells to proliferate,migrate,and invade.Conclusion SNW1 ha
