摘  要:目的:建立一种快速、简便、准确鉴别牡丹、芍药及川赤芍的DNA特征序列(DSS)标记分子鉴定方法。方法:基于芍药属植物叶绿体基因组数据库,采用IdenDSS软件筛选出DSS组及其特异性引物对,经DNA提取、聚合酶链式反应扩增、测序、序列比对进行牡丹、芍药及川赤芍目标DSS验证。结果:通过对5对候选DSS的验证,确定联合2个DSS(GTAACTTGCTATCCTCTTGCCTAGCAGGCAAAGATTTACT、ACTACGAA GAAGTTCCGGGGGTTACAAAGGAAACTTCGAG)标记可用于牡丹、芍药及川赤芍的鉴定,将DSS上传至中药分子鉴定平台(http://www. herbsdna. com)标准数据库中,结合数据库的BLAST功能,可实现牡丹、芍药及川赤芍的快速、准确鉴定。结论:筛选的DSS标记可用于牡丹皮、白芍及赤芍的基原鉴定。Objective:To establish a quick,convenient,and accurate DNA signature sequence tag(DSS)-based molecular identification method to identify Paeonia suffruticosa,P.lactiflora,and P.veitchii.Methods:DSS groups and their specific primers were screened by using IdenDSS software based on the chloroplast genome database of plants in Paeonia L..After DNA extraction,polymerase chain reaction(PCR)amplification,sequencing,and sequence alignment,the target DSSs used to validate P.suffruticosa,P.lactiflora,and P.veitchii were obtained.Results:The verification results of five candidate DSSs determined that two DSSs(GTAACTTGCTATCCTCTTGCCTAGCAGGCAAAGATTTACT and ACTACGAAGAAGTTCCGGGGGTTACAAAGGAAACTTCGAG)could be used for the identification of P.suffruticosa,P.lactiflora,and P.veitchii.The DSSs were uploaded to the Traditional Chinese Medicine Molecular Identification Platform(http://www.herbsdna.com).With the help of the BLAST function,a rapid and accurate identification of P.suffruticosa,P.lactiflora,and P.veitchii was achieved.Conclusion:The screened DSSs could be effectively applied to the origin identification of P.suffruticosa,P.lactiflora,and P.veitchii.
