摘  要:目的:优选关节止痛汤的提取工艺。方法:在单因素考察的基础上,以加水量、提取时间、提取次数为考察因素,以京尼平苷酸、松脂醇二葡萄糖苷、阿魏酸、杯苋甾酮的含量和出膏率为评价指标,采用层次分析法(AHP)-熵权法确定各指标权重。通过正交试验和遗传算法(GA)-反向传播(BP)神经网络法优选关节止痛汤的提取工艺参数,并对2种方法所得工艺参数进行验证比较。结果:正交试验所得最佳工艺参数为加水量6倍、提取时间0.5 h、提取3次,综合评分为90.21(RSD为1.38%);GA-BP神经网络优化得到的最佳工艺参数为加水量6倍、提取时间1.5 h、提取4次,综合评分为99.26(RSD为0.09%),结合实际生产需求,最终确定关节止痛汤的最佳提取工艺参数为加水量6倍、提取时间0.5 h、提取3次。结论:采用正交试验结合GA-BP神经网络所优选的提取工艺参数稳定、可靠,可为后续研发提供参考。Objective:To optimize the extraction process of Guanjie Zhitong Decoction.Methods:On the basis of single factor investigation,the water amount,extraction time,and extraction times were taken as the factors to be optimized.With the content of geniposidic acid,pinoresinol diglucoside,ferulic acid,and cyasterone and the extraction yield as evaluation indicators,the analytic hierarchy process(AHP)-entropy weight method was employed to determine the weight coefficient of each indicator.The extraction process of Guanjie Zhitong Decoction was optimized by the orthogonal test and genetic algorithm(GA)-backpropagation(BP)neural network method.The process parameters obtained with the two methods were analyzed and compared.Results:The optimal process parameters obtained by the orthogonal test were extraction with 6 volumes of water for 3 times(each of 0.5 h),which showed the comprehensive score of 90.21 points(RSD=1.38%).The optimal process parameters obtained by the GA-BP neural network method were extraction with 6 volumes of water for 4r times(each of 1.5 h),which showed the comprehensive score of 99.26 points(RSD=0.09%),slightly higher than that of the orthogonal test.Considering the actual production demand,it was finally determined that the optimal extraction process of Guanjie Zhitong Decoction was extraction with 6 volumes of water for 3 times(each of 0.5 h).Conclusion:The extraction process optimized by orthogonal test combined with GA-BP neural network was stable and reliable,which can provide reference for subsequent research and development.
