北美香柏精油提取工艺优化研究

摘  要:北美香柏树形优美、耐修剪、香气独特,是优良的观赏植物和重要的香料植物。其枝叶提取的精油具有杀菌功效、可入药,具有广阔的市场应用前景。为了进一步推动北美香柏精油的开发利用,以北美香柏鲜叶为试材,在单因素试验基础上,选取氯化钠浓度、浸泡时间和蒸馏时间3个对出油率有显著影响的因素,再结合Box-Behnken响应面法,对北美香柏精油提取工艺进行优化。结果表明:随着氯化钠质量浓度增加,出油率呈先增后减的趋势,当氯化钠质量浓度为0.04 g·mL^(-1)时,出油率最高为1.38%。随浸泡时间延长,出油率呈先上升后下降的趋势,浸泡16 h出油率达到峰值。随着蒸馏时间延长,精油提取率先升高后逐渐降低,蒸馏2 h的提取率最高。经过响应面试验,确定北美香柏精油的最佳提取工艺为氯化钠质量浓度0.043 6 g·mL^(-1)、浸泡16.4 h、蒸馏2.1 h,实际出油率为1.75%。The Thuja occidentalis boasts an elegant tree form,tolerance to pruning,and a unique aroma,making it an excellent ornamental plant and an important spice plant.The essential oil extracted from its branches and leaves possesses antibacterial properties and can be used as a medicinal ingredient,indicating broad market application prospects.To further promote the development and utilization of Thuja occidentalis essential oil,fresh leaves of the plant were used as test materials.Based on single-factor experiments,the extraction process of Thuja occidentalis essential oil was optimized using the Box-Behnken response surface methodology.The results showed that as the sodium chloride concentration increased,the oil yield first increased and then decreased,peaking at 1.38%when the sodium chloride concentration was 0.04 g·mL^(-1).With prolonged soaking time,the oil yield initially rose and then declined,reaching a peak after soaking for 16 hours.As the distillation time increased,the extraction rate of essential oil first rose and then gradually decreased,with the highest extraction rate observed after distillation for 2 hours.After the response surface experiment,the optimal extraction process was determined to be a sodium chloride concentration of 0.0436 g·mL^(-1),soaking for 16.4 hours,and distillation for 2.1 hours,resulting in an actual oil yield of 1.75%for the Thuja occidentalis.
