摘 要:目的阐明济川煎干预慢传输型便秘(slow transit constipation,STC)大鼠与ERK2/JNK1信号通路的关系。方法采用2018年11月课题组研究济川煎对STC模型大鼠肠道水通道蛋白的影响研究所保存的结肠组织标本。采用Western Blot、免疫组化检测及mRNA-实时荧光定量检测结肠组织中AKT1、TP53、MAPK1(ERK2)、MAPK8(JNK1)的表达水平;结果采用IBM SPSS 26.0软件进行统计分析,P<0.05时表示差异有统计学意义。结果济川煎可显著抑制STC模型大鼠结肠组织中AKT1、TP53、MAPK1、MAPK8的表达水平。结论济川煎可通过调控ERK/JNK信号通路来抑制STC模型大鼠结肠组织中AKT1、TP53、MAPK1、MAPK8的表达水平来发挥便秘的治疗作用。Objective To elucidate the relationship between Jichuan decoction and the ERK2/JNK1 signaling pathway in STC rats.Methods In November 2018,the research group studied the effect of Jichuan decoction on intestinal aquaporins in STC model rats,and preserved colon tissue samples were analyzed.The expression levels of AKT1,TP53,MAPK1(ERK2),and MAPK8(JNK1)in colon tissues were quantitatively detected by Western Blot,immunohistochemical detection,and mRNA-real-time fluorescence.Results IBM SPSS 26.0 software was utilized for statistical analysis,with P<0.05 indicating a statistically significant difference.Results Jicuan decoction can significantly inhibit the expression levels of AKT1,TP53,MAPK1,and MAPK8 in the colon tissue of rats with slow transit constipation(STC)model.Conclusion By regulating the ERK/JNK signaling pathway,Jichuan decoction promotes ICC growth,increases SP and VIP,AQP3 and AQP4 expression levels,improves intestinal flora,and promotes intestinal endothelial cell repair.
