摘 要:建立高效液相色谱法测定植物提取物及其制品中γ-氨基丁酸含量。该研究通过对比7种衍生剂对植物提取物及其制品中γ-氨基丁酸测定的影响来确定适用的衍生剂,并优化衍生条件和色谱条件,再依次对提取溶剂、料液比、提取时间、提取次数开展单因素实验。该方法采用水提、6-氨基喹啉基-N-羟基琥珀酰亚胺基氨基甲酸酯(AQC)衍生,衍生物用Agilent ZORBAX Eclipse Plus C18色谱柱分离并以磷酸盐缓冲液-乙腈体系等度洗脱,采用二极管阵列检测器检测,检测波长248 nm,外标法定量。该方法线性范围为2.00 mg·L^(-1)~100 mg·L^(-1)(R2=1.0000),检出限为0.67 mg·kg^(-1),定量限为2.23 mg·kg^(-1),回收率85.5%~104.9%。该方法污染小、毒性低、检测试剂易获取、检测成本较低廉,具有良好的准确度、重现性和稳定性,可满足大批量样品检测需求,对γ-氨基丁酸功能性食品开发及质量控制有积极意义。To construct a high performance liquid chromatographic method for the determination of y-aminobutyric acid in plant extracts and their products.In this study,the effects of seven derivatization reagent on the determination of y-aminobutyric acid in plant extracts and their products were compared to determine the suitable reagent and optimise the derivatisation conditions and chromatographic conditions,and thensingle factor experiment were carried out on the extraction solvents,the material-liquid ratio,the extraction time and the number of extractions in turn.Results The method was based on aqueous extraction,derivatisation with 6-aminoquinolinyl-N-hydroxysuccinimidyl carbamate(AQC),and the derivatives were separated and isocratic eluted with phosphate buffer-acetonitrile system on an Agilent ZORBAX Eclipse Plus C18 column,and detected by a diode array detector at a detection wavelength of 248 nm and quantified by external standard method.The linear range of the method was 2.00 mg·L^(-1)~100 mg·L^(-1)(R²=1.0000),the detection limit was 0.67 mg·kg,the quantification limit was 2.23 mg·kg,and the recoveries were 85.5%~104.9%.The method is of low contamination,low toxicity,easy to obtain detection reagents,low cost,and has good accuracy,reproducibility and stability,which can meet the demand of large-volume sample detection and has positive significance for the development and quality control of y-aminobutyric acid functional foods.
